Genetic Structure and Diversity of Native Tench (Tinca tinca L. 1758) Populations in Hungary—Establishment of Basic Knowledge Base for a Breeding Program

Tench is a cyprinid fish that has undergone human-induced translocations. The natural populations of the species are on the decline due to habitat loss and spawning grounds degradation. The genetic diversity of seven natural populations was investigated to establish the genetic knowledge base for successful conservation efforts and for selective breeding. Twelve microsatellite markers, the sequencing of a 615 bp section of mtDNA (Cytb) and PCR-RFLP analysis of two nuclear markers (Act) and (RpS7) were used to analyze the genetic variation and structure among 175 individuals. Randomized complete block design with three replicates was used to study the general combining ability of parents, and specific combining ability of 12 F1s. Combining ability was studied in a partial diallel cross in maize for traits yield and attribute at two locations (Qlyasan and Kanipanka) at Sulaimani Governorate. All amplicons were well suited for the prepared primer with size 873 bp . and identical ratio ranged 87.2- 98% of the same species, as included a high similarity of the isolates taken camels in Iran and Iraq related to Parabronema skrjabini . Phylogenic tree inferred the degree of relatedness between 28SrDNA sequences deposited in the international nucleotide bank sequence database (NCBI). The sequence of Parabronema skrjabini was recorded in the Genbank under accession number MT742154.1.

Forty five sites were randomly sampled and analyzed for hierarchical cluster analysis which has been used to group samples by using Ward's method and to develop soil maps. The spatial distribution of distinct groups of elements demonstrates the interplay of ECe, Gypsum content, and particle size distribution factors. This study was aimedto evaluate the influence of carrageenan and pectin on jam color during 10 days of cold storage (+ 4 ºC). Jam samples were prepared with cherry and apricot fruits and 3 concentrations of gelling agents (0.5 %, 1 % and 2 %). Cherry and apricot jam samples prepared without gelling agent were used as control samples. Pectin, κ-carrageenan and ι-carrageenan were used as gelling agents. The jams were stored in the refrigerator for 10 days.
