Detection of Human Cytomegalovirus in Colorectal Adenocarcinoma by In Situ Hybridization Technique

Background: Human Cytomegalovirus (HCMV) infects a wide range of human cells, including colonic epithelial cells that give rise to adenomas and adenocarcinomas. Persistent productive infection of tumor cells is essential for oncomodulation by HCMV.This study aimed to detect HCMV matrix protein using in situ hybridization technique (ISH) in colorectal adenocarcinoma compared to normal colon tissues, and to the presence of cytomegalovirus inclusion bodies in infected colorectal carcinomas.
Patients and methods: Twenty six of colorectal adenocarcinomas were obtained in paraffin-blocks compared to 10 normal colon specimens which were age and sex matched as control group. Detection of HCMV was obtained by in situ hybridization technique.
Results:The biotinylated probe specific for DNA encoded HCMV-matrix protein showed hybridization with nucleic acid in 20 cases out of (26) of colorectal adenocarcinomas representing (76.9%) compared to normal colon tissue which revealed no hybridization signals. Moderate to high scoring signals were detected in moderate to poorly differentiated groups. Inclusion bodies were detected in 11 (42.3%) cases with positive hybridization signals.
Conclusion: HCMV may play a role in the colorectal adenocarcinoma pathogenesis. In situ hybridization test are considered the most sensitive and specific tools for detection of HCMV DNA in tissues. Epidemiological, histopathological identification of cytomegalic inclusion bodies and molecular studies are necessary to confirm the association of HCMV and colorectal tumorogenesis in Iraqi population.

Detection of Human Parvovirus B19 antibodies in Pregnant Women with Spontaneous Abortion

Background: Viral infection in pregnancy is known to cause many problems such as fetal malformation; growth restriction and even fetal death; the relationship between viral infection and abortions is not well understood. The infection during pregnancy can affect the fetus due to lack of mother’s immunity.
Objective: To assess the frequency of human parvovirus B19-specific antibody type IgM and IgG among pregnant women with spontaneous abortion.
Patients and Methods: A semi-quantitative enzyme linked immunosorbent assay technique was applied for detection of anti-human parvovirus B19 IgM (Ridascreen-K6031-Germany) and IgG (Ridascreen -K6021- Germany) in sera of 90 pregnant women who have previously experienced abortion were collec

Bioequivalence of Two Formulations of Amoxicillin in Human Healthy Volunteers on (HPLC) Technique

Amoxicillin is commercially available in the form of capsules and tablets containing 250mg or 500mg for oral administration. It is also available in the form of suspension containing "25mg/ml” . Amoxicillin is presently used as the most common antibiotics .Ten healthy Human volunteers were characterized respected to their pharmacokinetic and bioavailability of two formulations of Amoxicillin from two sources of  industrial companies  after a single dose administration was given orally. A procedure is described for determination the concentration levels of Amoxicillin in human plasma of healthy volunteers using high performance liquid chromatography (HPLC) with reversed-phase isocratic column at low wave length of UV-visib

Molecular Detection of High-Risk Human Papillomavirus Genotypes from Cervical Lesions in Baghdad

Objective Using two complementary techniques of virus detection human papillomavirus (HPV)[capture of hybrids (CH) and polymerase chain reaction (PCR)], relate the cytological study and/or cervical biopsy with high-risk HPV (HPV-HR) genotypes presence, as well as relating their viral load (VL). Methods About 272 women, who presented most cell alterations compatible with lesions cervical HPV, which has been detected in all high risk by the CH method and HPV genotype detection by PCR. Results In 22% of the patients it was not detected HPV DNA. Genotype 16 and/or 18 was prevalent and was found in 33% of the 212 women studied, meanwhile, mixed infections were found by several genotypes in 25%. In as for the histological lesions found, in 61 pat

Detection of Biofilm Genes (gtf) in Streptococcus mutans Isolated from Human Dental Caries

Sixty samples from saliva and dental plaque were selected from patients with caries active at ages from 4-65years. 22 isolates belong to Streptococcus mutans. All isolates pronounced adhesion and biofilm formation in various degrees. By using Polymerase Chain Reaction ﴾PCR﴿ Techniques, it was found that these isolates had gtfB encode GtfB with 80 bp, gtfC encode GtfC with 81 bp, and gtfD with 324 bp which explain their potential of biofilm formation.

Detection of Single Nucleotide Polymorphisms (SNPs) for Genes Cause Drug-Resistant in Iraqi Mycobacterium Tuberculosis isolates by new Pyrophosphate Technique.

In this search, a new pyrophosphate technique was proved. The technique was employed to single- nucleotide polymorphisms (SNPs), which diagnosis using a one-base extension reaction. Three Mycobacterium tuberculosis genes were chosen (Rpob, InhA, KatG) genes. Fifty-four specimens were used in this study fifty-three proved as drug-resistant specimens by The Iraqi Institute of Chest and Respiratory Diseases in Baghdad.; also one specimen was used as a negative control. The steps of this technique were by used a specific primer within each aliquot that has a short 3-OH end of the base of the target gene that was hybridized to the single-stranded DNA template. Then, the Taq polymerase enzyme and one of either α-thio-dATP, dTTP, dGTP, or dCTP

Misleading Presentation of Colorectal Cancer in an Otherwise Healthy Patient

Background: Colorectal cancer (CRC) represents the second most common malignancy and the fourth most common cause of cancer deaths. CRC can manifest early with bright red bleeding per rectum, tenesmus, and altered bowel habits. These symptoms are often attributed to benign lesions, including anal fissure. Our objective is to highlight the alarming scenario of an anal fissure masking the clinical features of an underlying colorectal cancer in healthy middle-aged patients.

Case Report

Our case report aims to discuss how congruent clinical features of benign-looking anal fissure can delay the diagnosis of rectal cancer. In January 2019, a healthy forty-four years old Iraqi male with no famil

The possible role of EBV in carcinogenesis of colorectal carcinoma.

Background: Except for the tight correlation to nasopharyngeal carcinoma, accumulating evidences show that Epstein-Barr virus (EBV) is correlated to other carcinomas. This study was to investigate the association of EBV with colorectal tumors.
Materials & methods: Forty paraffin embedded blocks of colorectal tumors (thirty were adenocarcinoma and ten were benign tumors) were all examined for the presence of EBV DNA with
the application of In Situ hybridization.
Results: In Situ EBV DNA signals was detected in 6 out of 30 (20%) of colorectal carcinoma with no observed signals in the sections from benign group.
Conclusion: Our results showed that infection of EBV exists in human colorectal adenocarcinoma

Seroprevalence of Toxoplasma gondii and Cytomegalovirus in Aborted Women in Baghdad-Iraq

Aim of this study was to assess the seroprevalence of Toxoplasmosis and
Cytomegalovirus (CMV) infection among aborted women. This study was
performed on 282 aborted women and 50 normal pregnant women (control group)
collected from Al-Yarmouk Teaching Hospital and Fatima Al-Zahraa Hospital for
Obstetrics and Children during October 2013 until the end of February 2014.The
samples were tested with Latex Agglutination Test (LAT) and CMV Rapid Test
Cassette as a primary screening for T. gondii and CMV antibodies in the women
sera. The primary tests showed a percentage of positive antibodies of T. gondii in
aborted women and normal pregnant women 99(35.1%), 13(26%) respectively,
while for CMV, it was 101(95.3%), 36(7
