Detection of Human Cytomegalovirus in Colorectal Adenocarcinoma by In Situ Hybridization Technique

Background: Human Cytomegalovirus (HCMV) infects a wide range of human cells, including colonic epithelial cells that give rise to adenomas and adenocarcinomas. Persistent productive infection of tumor cells is essential for oncomodulation by HCMV.This study aimed to detect HCMV matrix protein using in situ hybridization technique (ISH) in colorectal adenocarcinoma compared to normal colon tissues, and to the presence of cytomegalovirus inclusion bodies in infected colorectal carcinomas.
Patients and methods: Twenty six of colorectal adenocarcinomas were obtained in paraffin-blocks compared to 10 normal colon specimens which were age and sex matched as control group. Detection of HCMV was obtained by in situ hybridization technique.
Results:The biotinylated probe specific for DNA encoded HCMV-matrix protein showed hybridization with nucleic acid in 20 cases out of (26) of colorectal adenocarcinomas representing (76.9%) compared to normal colon tissue which revealed no hybridization signals. Moderate to high scoring signals were detected in moderate to poorly differentiated groups. Inclusion bodies were detected in 11 (42.3%) cases with positive hybridization signals.
Conclusion: HCMV may play a role in the colorectal adenocarcinoma pathogenesis. In situ hybridization test are considered the most sensitive and specific tools for detection of HCMV DNA in tissues. Epidemiological, histopathological identification of cytomegalic inclusion bodies and molecular studies are necessary to confirm the association of HCMV and colorectal tumorogenesis in Iraqi population.

Detection of Cytomegalovirus, Rubella virus, and IL-2 Levels in a Sample of Recurrently Aborted Iraqi Women

The present study aimed to investigate CMV and Rubella virus as a causative agent of recurrent abortion, while the IL-2 levels were estimated as immune parameter during pregnancy period. A total of 63 blood samples were collected from recurrently aborted women, control non-pregnant women and control pregnant women. The results recorded 72.09 % CMV positive aborted women and 27.91 % Rubella virus positive aborted women. Levels of IL-2 were (437.03 ± 38.02) pg/ ml in first group, (390.51± 63.56) pg/ ml in second group, (32.98 ±15.12) pg/ ml in control group non pregnant women and (118.63 ± 24.81) pg/ ml in control pregnant women. High IL-2 levels in all studied women indicate presence of a factor affecting the immune system other than

Detection of Anti-rubella virus, Cytomegalovirus and Chlamydia pneumonia antibodies in patients with type I diabetes mellitus

Type-1 diabetes is defined as destruction of pancreatic beta cell, virus and bacteria are some environmental factor for this disease. The study included 25 patients with type-1 diabetes mellitus aged between 8 – 25 years from Baghdad hospital and 20 healthy persons as control group. Anti-rubella IgG and IgM, anti-Chlamydia pneumonia IgG and IgM were measured by ELISA technique while anti-CMV antibody were measured by immunofluorescence technique. The aim of current study was to know the trigger factor for type-1 diabetes. There were significant differences (P<0.05) between studied groups according to parameters and the results lead to suggest that Chlamydia pneumonia, CMV and rubella virus may trigger type-1 diabetes mellitus in Iraqi pat

Predicting maize ear grain weight in situ by ear dimensions

To find out a simple and efficient equation to estimate maize ear grain weight on farm (in situ), twenty three maize crosses along with two synthetics were grown in the field. On the experimental farm of the Dept. of Field Crop Sci., College of Agric., Univ. of Baghdad, seeds of twenty five maize genotypes were grown in the fall season of 2013 with three replicates. At dough stage of the kernels, five naked ears of each experimental units were measured for length and maximum diameter. This will sum up 125 ears of the trial. The volumes of ears were calculated as cylinder (length× r2× 3.1416). Grain weight of all ears were determined after harvesting and drying to 15% grain moisture. A constant was calculated by dividing ear grain weight b
