Phylogenetic tree analysis based on the 16S sequence alignment for Klebsiella spp. isolated from different sources

16S ribosomal RNA (16S rRNA) gene sequences used to study bacterial phylogeny and taxonomy have been by far the most common housekeeping genetic marker utilized for identification and ancestor determination. This study aimed to investigate, for the first time, the relationship between Klebsiella spp. isolated from clinical and environmental samples in Iraq.

     Fifty Klebsiella spp. isolates were isolated from clinical and environmental sources. Twenty-five isolates were collected from a fresh vegetable (Apium graveolens) and 25 from clinical samples (sputum, wound swab, urine). Enteric bacteria were isolated on selective and differential media and identified by an automatic identification system, vitek-2. The total DNA was extracted and PCR amplified for selected isolates. The 16S rRNA gene was amplified by using the universal primer 27F (5'- AGAGTTTGATCCTGGCTCAG- 3') and 1492R (5'- GGTTACCTTGTTACGACTT- 3’). The 16SrRNA gene sequence was analysed among some local isolates, and the results were compared with the standard data of similar registered strains in NCBI. The most common species of Klebsiella was Klebsiella pneumoniae pneumoniae (Kpp), followed by Klebsiella pneumoniae ozaenae (Kpo) and Klebsiella oxytoca (Ko). The results of the identification of species and sub species by using the  biochemical test (vitek-2) were more precise than those obtained by the use of the universal primer.Phylogenetic tree strategies have clearly indicated a relatively close similarity amongst all analysed Klebsiella isolates and revealed the intra-species genetic distance between the individual isolates of the Klebsiella spp. In conclusion, our results revealed the main advantage of using universal primers for the identification of Klebsiella spp. and their root from nature.

ABSTRACT The isolation and characterization of (27) isolate of extreme halophilic bacteria was performed ninteen isolate belonged to the genus Halobacterium which included Hb.halobium. Hb. salinarium, Hb. volcanii. Growth curve and generation time in logarthmic phase was measured and found to be (12.8hr±0.32), (11.2hr±0.2), (9.8hr±0.87), respectivaly. Effect of various concentrations of NaCl, KCI, NH4Cl and MgSO4.7H2O was studied, NaCl was essential for the rod shape rapid growth Rat and pigmentation. Less than 1% concentration caused lysis of bacteria. Yeast extract was the best carbone source as compared with glucose and casamino acid.

Effect of lead on biofilm formation by environmental isolates of Bacillus spp.

Using ISSR markers to build a phylogenetic of Barley Genotypes

This study is attempts to build a phylogenetic between nine Iraqi barley
genotypes based on ISSR-PCR analysis by determine the level of genetic similarity
among them. Nine issr primers used in this study produced 41 bands across nine
studied varieties. Of these bands, 28 bands were polymorphic and the remaining
monomorphic bands were 13. The average polymorphic rate was 70.5% ranged
between 25%-100% , and average of polymorphic bands /primer was 4.5.The size
of the amplified bands ranged 140-1600 bp. It was generated a 5 unique bands in
this study, these bands can be used as a DNA profiling of all studied genotypes. The

The Effect of Alcoholic Extracts of Zingiberofficinale Anti-E.Coli Isolates Isolated from Urinary Tract Infection

Synergistic Effect of Linezolid, Tigecycline, and Vancomycin on Staphylococcus Aureus Isolated From Iraqi Patients with Diabetic Foot Ulcers

Ninety nine swabs were collected from patients with diabetic foot ulcers (DFU), all swabs   were cultured on different selective media for screening, 46 isolates confirmed as S. aureus by API staph. The results of antibiotic susceptibility test revealed that all isolates were resistant to metronidazole, 34 isolates were resistant to cefoxitin, ceftriaxone, and meropenim, 23 isolates were resistant to ciprofloxacin and norfloxacin, 17 and 16 isolates were resistant to tetracycline and trimethoprim, respectively; while all isolates were sensitive to tigecycline.
