Optimum Condition of Inulinase Production from Bacillus cereus (Be9)

Fifty isolates of Bacillus spp were obtained from rhizosphere soil of compositae
plant roots. The ability of inulinase production by these isolates was screened.
Bacillus Be9, which isolated from soil of lettuce root, was the highest inulinase
producer; it was identified as Bacillus cereus. Optimal culture medium and
condition for inulinase production were determinatd; the highest inulinase
production was obtained when the bacteria was cultured in inulin medium which
contained 0.5% inulin, 0.4% peptone as carbon and nitrogen source at pH 7.0
inoculated with 1ml of bacterial suspension and incubated at 40˚C for 48hrs.

Biofuel Production from Cellulosic Wastes by Local Isolates of Streptomyces sp.

The current study aims to produce cellulase enzyme from Streptomyces spp. isolates and study the effect of some cultural conditions on cellulase production; biofuel production from cellulotic waste through enzymatic and acids hydrolysis. Out of 74 isolates of Streptomyces sp. were screened for cellulse production in solid and liquid media. Results showed higher capability of isolate Streptomyces sp. B 167 for cellulase production and bioconversion of cellulose, therefore selected for further studies. The results of optimization revealed that the cellulase enzyme productivity by the selected isolate reached 2.1 and 2.28 U/ml after 48 h of incubation time and pH 7 respectively. Cellulase productions in tested isolate improved (2.57 U/ml) b

Improvement Efficacy of Bacillus Subtilis Cellulose Hydrolyzing by Using Cold Plasma Technique

Bacillus subtilis, an isolate of bacillus genus, was obtained from the laboratories of Ministry of Science and Technology. The best efficient Bacillus subtilis isolate in cellulose and semi-cellulose hydrolysis was treated with Dielectric-barrier Discharge (DBD). Atmospheric cold plasma technique (non-thermal) was used by exposing them at different times (2, 3, 4 and 5 mins) separately as a first stage, and then 60 seconds after any treatment separately as a second stage. After 48 hours, the difference between the plasma source and the sample was fixed at 0.5 cm. The results showed a variation in the growth of the isolate according to the exposure time by the appearance of culture turbidity and the estimation o

Optimum Height of Plate Stiffener under Pressure Effect

The economical design of plate loaded by pressure can be obtained by using stiffeners instead of increasing the thickness of plate. The main subject of this work is to obtain the effect of stiffener height on the maximum stress in the plate subjected to pressure load. Different plate-stiffener sets are selected to find the effects of stiffener thickness, plate dimensions and pressure, on the optimum stiffener height. The models under consideration are square plates clamped rigidly from four edges. Finite Element method is used to analyze 160 different models by using the Finite Element software package ANSYS version 11. Another analysis method based on maximum stress equation is used to analyze 30 models. The graphical comparison of results

Bioethanol Production from Olive Solid Residues by Using Rhodotorula Minuta

Bioethanol is an attractive fuel with higher potential for energy security and environmental safety. Olive solid residues were used as a raw material for the production of bioethanol through the use of different preliminary treatments . Separate treatments with cellulose, hydrochloric acid (HCl 5%), sulfuric acid (H2SO4 2%), and liquid ammonia NH4OH (20%) were used to convert cellulose and hemicellulose into monosaccharaides. The production of ethanol was observed during the fermentation process using R. minuta under anaerobic conditions.  After 3 days of fermentation, lowest concentrations of ethanol of  0.233, 0.249, 0.261, and 0.275 g/ l were produced from ol

Production, Purification and Characterization of Cellulose from Local Isolate of Pantoea spp

Soil samples from fields cultivated with barley and wheat in addition to samples
from spoiled orange and apple fruits and carrot roots were collected with the aim to
isolate cellulase producing bacterial strains. Bacterial isolates obtained from these
samples were grown on a selective medium containing carboxymethyl cellulose
(CMC) as a sole source for carbon and energy. Results showed that nine isolates out
of fifty were able to produce cellulase. The specific activity of cellulase in culture
filtrate of the most efficient isolate was 1.601 u/mg protein. This isolate was
identified according to its morphological characteristics and biochemical tests, and
then by using Api 20-E and VITEK-II identification systems an

The Production of Ethanol from Sugar Beet Waste by Immobilized Saccharomyces Cerevisiae

Saccharomyces Cerevisiae cells were immobilized in calcium alginate beads and activated charcoal for use in the
production of ethanol from batch fermentation of sugar beet waste. Treatment of the waste with NaOH to increase the
ability of lignocellulose material to hydrolysis by acid (2N H2SO4) to monosaccharide and disaccharide (mainly glucos).
The high reducing sugar concentration obtained was equal to 9.2gm/100ml (10Brix) after treatment. Fermentation
parameters, are (pH, glucose concentration (2.5-25 gm/100ml), immobilized agent concentration (2.5-25 gm/100ml)
were studied to find the optimum physiological condition. And the highest ethanol concentration obtained from the
fermentation in the presence of 20%(wt/v) ca

Optimization of Bis-anthraquinones Production from Endophytic Fungi Diaporthe sp. GNBP-10

Bis-anthraquinones with a unique molecular backbone, (+)-2,2’-epicytoskyrin A (epi) and (+)-1,1′-bislunatin (bis), was produced by endophytic fungi Diaporthe sp GNBP-10 associated with Gambir plant (Uncaria gambier). Epi and bis possess robust antimicrobial activity toward various pathogens. This study focus on knowing the optimum condition of epi and bis production from Diaporthe sp GNBP-10. A series of culture media with various nutrient compositions was investigated in epi and bis production. The content of epi and bis was determined by measuring the area under the curve from TLC-densitometric (scanner) experiment. The linear regression analysis was then applied to obtain the results. The optimi

Production of antibacterial agent from Streptomyces griseus by using Semi Solid Fermentation

Backround: The Solid state fermentation has several advantage including absence of free water , reduced volume of production media utilized for high products and the relatively low costs of production.
Methods: Thirty local isolates of soil obtained from Genetic Engneering and Biotechnology Institute. Nutrient agar was used to growth strains examination to antibacterial agent and Wheat bran and fish meal were used in combination (0-100%of each )and divided in 10 gm lost /flask . Each flask is inoculated with different numbers of Streptomyces spores and incubated for 5 days at 28°C, then the supernet was extracted and were assayed as antibacterial
Results: The ability of 30 local isolates of Streptomyc
