Introduction: Melanin is a high-molecular weight pigment produced through the oxidative polymerization of phenolic or indolic compounds and plays a perfect role in UV-light shielding, as well as in photoprotection. Among biopolymers, melanin is unique in many aspects. This study is designed to screen Production, extraction and characterizes of an extracellular melanin pigment from clinically isolated P. aeruginosa. Objective: The aim of the current study is isolation and diagnosis of P.aeruginosa using vitek-2 compact system and screening the ability to produce melanin and characterization of extracted melanin by UV-vis, FTIR, XRD and SEM. Materials and methods: the samples swab inoculated on cetrimide agar as selective media and incubated aerobically for 24 hours at 37°Cand used nutrient agar with nutrient broth supplement with 1% tyrosine for Screening for melanin production by P. aeruginosa isolates. Results: Four isolates P. aeruginosa were identified out of 109 specimens have ability to produce melanin pigment under specific medium and culture conditions. According to morphological, cultural, biochemical, and VITEK-2 characteristics, isolates were identified as P. aeruginosa. The results showed that the isolate of burn was greatest in production of melanin. Based on its solubility in organic solvents, the black pigment was identified as melanin and structurally and functionally characterized by UV-Vis absorbance and presence of various characteristic peaks that determined by FTIR analysis, and the morphology of the pigment surface was examined using SEM and XRD spectra analysis. Conclusion: Depending on the type of bacteria and the conditions of their culture, different melanin-producing bacteria produce different amounts of melanin. The primary factors for bacterial melanin production are L-tyrosine, energy source, pH, temperature and surface area for shaker agitation. In this study, results found that the optimum condition for melanin production byP. aeruginosa in pH=7.3 and the effect of L-tyrosine concentration on melanin production found that use 1g/L is the best concentration for production.
This study was conducted to isolate and identify killer yeast Hanseniaspora uvarum from dates vinegar and measurement the ability of this yeast to produce killer toxin. The antimicrobial activity of the concentrated supernatant containing partially purified concentrated killer toxin was also detected against several pathogenic bacteria and yeast species, which includes two types of yeast Rhodotorula mucilaginosa and Candida tropicalis and four human pathogenic bacteria Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeurginosa. In addition, the antagonistic activity of examined yeast have been studied toward four types of fungi, where two are pathogenic

Microorganisms have an active role in biotechnology for example yeasts, especially in some genus like Saccharomyces, Pichia, and Candida. C.tropicalis one of the most important species of Candida and despite it is one of the causative agents of candidiasis but it has a major role in the production of many chemical compounds. C.tropicalis in the previous study was isolated from sheep dung and morphologically and molecularly classified the result of sequencing was elucidate 100% similarity between the studied isolate and other isolates inserted in DNA Data Bank of Japan DDBJ, physiologically this isolate tolerated 6% ethanol concentration in broth media with the ability to the pro

The inhibitory effect of partially purified LPS on Candida glabrata yeast was studied in a microdilution method. This study found that LPS has an inhibitory effect on Candida glabrata with the lower concentrations. The inhibitory effect of LPS which treated with heating was studied under boiling and wet heat effect. The toxicity of LPS on Candida glabrata was not affected when treated with heating LPS and the results were similar to those found in untreated LPS
Forty-five samples in total there are just twenty isolates of Pseudomonas aeruginousa. Over the period of four months, from January 2025 to April 2025, a variety of clinical samples were gathered from numerous hospitals in Baghdad, including Al-Sader, Ibn Al-Balady, Fatima Al-Zahraa, and Al-Imam Ali Hospitals. The clinical samples of patients with burns infections were included. The Vitek 2 compact and a biochemical test verified that all isolates were cultivated on king A, king B, ctrmide agar, macConkey agar, and blood agar. Using the disk diffusion technique, the susceptibility of P. aeruginousa isolates to 16 antibiotics from various classes was determined. P. aeruginousa isolates exhibited strong resistance to the majority of t

The ability of the bacterial species to form biofilm onto polyvinyl chloride (PVC) is associated with several economic, health and environmental problems. The effect of kind of water on ability of this bacterium to form biofilm is scanty in literature. In present study, the ability of different environmental isolates of P. aeruginosa to form biofilm onto polystyrene microtiter plate was evaluated. Furthermore, the effect of waters that collected from different sources on biofilm formation of this bacterium onto PVC was studied. Spectrophotometric method was used to check the ability of bacteria to form biofilm and evaluated the role of waters onto a

Alginate, Psl and Pel are three exopolysaccharides that constitute the main components in biofilm matrix, with many biological functions attributed to them, especially concerning the protection of the bacterial cell from antimicrobial agents and immune responses. A total of 25 gentamicin-resistant P. aeruginosa selected isolates were enrolled in this study. Biofilm development was observed in 96% of the isolates. In addition, the present results clarified the presence of pelA and pslA in all the studied isolates. The expression of these genes was very low. Even though all biof
