Introduction: Melanin is a high-molecular weight pigment produced through the oxidative polymerization of phenolic or indolic compounds and plays a perfect role in UV-light shielding, as well as in photoprotection. Among biopolymers, melanin is unique in many aspects. This study is designed to screen Production, extraction and characterizes of an extracellular melanin pigment from clinically isolated P. aeruginosa. Objective: The aim of the current study is isolation and diagnosis of P.aeruginosa using vitek-2 compact system and screening the ability to produce melanin and characterization of extracted melanin by UV-vis, FTIR, XRD and SEM. Materials and methods: the samples swab inoculated on cetrimide agar as selective media and incubated aerobically for 24 hours at 37°Cand used nutrient agar with nutrient broth supplement with 1% tyrosine for Screening for melanin production by P. aeruginosa isolates. Results: Four isolates P. aeruginosa were identified out of 109 specimens have ability to produce melanin pigment under specific medium and culture conditions. According to morphological, cultural, biochemical, and VITEK-2 characteristics, isolates were identified as P. aeruginosa. The results showed that the isolate of burn was greatest in production of melanin. Based on its solubility in organic solvents, the black pigment was identified as melanin and structurally and functionally characterized by UV-Vis absorbance and presence of various characteristic peaks that determined by FTIR analysis, and the morphology of the pigment surface was examined using SEM and XRD spectra analysis. Conclusion: Depending on the type of bacteria and the conditions of their culture, different melanin-producing bacteria produce different amounts of melanin. The primary factors for bacterial melanin production are L-tyrosine, energy source, pH, temperature and surface area for shaker agitation. In this study, results found that the optimum condition for melanin production byP. aeruginosa in pH=7.3 and the effect of L-tyrosine concentration on melanin production found that use 1g/L is the best concentration for production.
The purpose of this study to synthesize and characterize silver nanoparticles using phenolic compounds obtained from Camellia sinensis, to test the antibacterial properties of biosynthesized nanoparticles on the formation of biofilms in multidrug-resistant Pseudomonas aeruginosa. Ten isolates of P. aeruginosa were obtained from the Genetic Engineering and Biotechnology Institute laboratories of the University of Baghdad. By using the VITEK-2 system and culturing the isolates on cetrimide agar, the diagnosis was confirmed. Camellia sinensis silver nanoparticles (CAgNPs) were created using an extract of the plant's aqueous and methanolic leaves. Based on the results of the nanoparticle synthesis, spherical nanoparticles that may be single or

Smith) were used in the current study to test their ability to production biodiesel through stimulated in different nitrogen concentration treatments (0, 2, 4, 8 gl ), and effect of nitrogen concentration on the quantity of primary product (carbohydrate, protein ), also the quantity and quality of lipid. The results revealed that starvation of nitrogen led to high lipid yielding, in C. vulgaris and N. palea the lipid content increased from 6.6% to 40% and 40% to 60% of dry weight (DW) respectively. Also in C. vulgaris, the highest carbohydrate was 23% of DW from zero nitrate medium and the highest protein was 50% of DW in the treatment 8gl. Bacterial isolates were identified using conventional biochemical tests and then identification was confirmed by using Vitek-2 compact system. Pseudomonas aeruginosa recovery was 60 isolates in this study. These isolates were analyzed for antibiotic susceptibility by the disk diffusion test (DDT) according to Kirby Bauer's method using seven clinically important antipseudomonal agents: carbapenems (Imipenem and Meropenem), pencillins (Piperacillin), cephalosporins (Ceftazidim), monobactam (Aztreonam), quinolones (Ciprofloxacin) and aminoglycosides (Gentamicin). Show MoreSiderophores are low molecular weight organic compounds produced by microorganisms growing under low iron concentration. In this study we describe the detection, production and extraction of siderophores secreted by Acinetobacter baumannii (Multiple-drug resistant ) pathogens. One hundered twenty Gram –negative non lactose fermenter bacilli isolates have been collected from three hospitals at Baghdad city over three months. Primary identification of these isolates is performed by standard diagnostic methods (biochemical tests and API 20 NE); 19 clinical isolates of A. baumannii are cultured on CHROMagar (highly selective medium for detection of MDR Acinetobacter) as well as diagnoses is documented by using Vitek 2 system. Isolates are exa

Their increase interest in creating and researching silver nanoparticles (AgNPs) because of their numerous applications in many fields especially medical fields such as burn, wound healing, dental and bone implants, antibacterial, viral, fungal, and arthropodal activities. Biosynthesis of nanoparticles mediated pigments have been widely used as antimicrobial agent against microorganisms. Silver nanoparticles had synthesized by using melanin from locally isolate Pseudomonas aeruginosa, and used as antimicrobial activity against pathogenic microorganisms. Aim of the study: Isolation of Pseudomonas aeruginosa

Show MoreThe present work aimed to investigate the neuraminidase (nan1) gene expression in 32 different clinical isolates of Pseudomonas aeruginosa to explore the role of the enzyme in different types of infection and might give a better understanding of host cell-pathogens interaction. In addition, the effect of monosaccharide D-mannose on neuraminidase gene expression in eight isolates was studied by utilizing a reverse transcription-quantitative polymerase chain reaction (RT-qPCR). The results demonstrated that the highest expression of nan1 gene was in otitis samples (208,913.81) which were significantly higher than that from other infections (P < 0.01). While, the concentrations of gene copies obtained from urin

Therefore, as a therapeutic option becomes restricted, the search for a new agent is a preference. So P. aeruginosa is an extremely versatile Gram-negative bacterium capable of thriving in a broad spectrum of environments, and this performs main problems to workers in the field of health. One hundred and fifty samples were collected from different sources from Baghdad hospitals, divided into two main groups: clinical (100) specimens and (50) samples as an environmental, collected from October 2019 to the March 2020. All of these samples were cultured by specific and differential
