Evaluation of The Genoprotective Effect of Curcumin Against Methotrexate in Bone Marrow and Spleen Cells in Mice

Curcumin is a yellow pigment produced from the rhizomes of the Curcuma longa plant and a primary chemo preventive component of turmeric is used as a spice and food coloring ingredient. Curcumin has a large number of pharmacological activities, such as anticancer, anti-diabetic, antioxidant, anti-infectious, and anti-inflammatory properties.Investigation of the geno-protective effect of curcumin on methotrexate induces chromosomal aberrations of spleen and bone marrow cells. In this study, 32 mice were used and divided into four groups (eight mice at each group) as follows: Group1 (negative control): Dimethyl sulfoxide was given intraperitoneally to mice every day for ten days.Group2 (positive control): Mice were received a single dose (20mg/kg) of methotrexate intraperitoneally Group3: Mice were received (200mg/kg) of curcumin solution intraperitoneally for ten successive days.Group4: Mice were received (200mg/kg) of curcumin solution intraperitoneally for ten successive days and on the day 11, a single dose  of methotrexate ( 20 mg/kg ) was injected intraperitoneally . In animals treated with methotrexate, significant elevations of the chromosomal aberrations were observed along with a decline in the mitotic index. Meanwhile, there was a considerable elevation of the mitotic index and no detectable chromosomal changes in the curcumin-supplemented mice. The number of abnormal cells was reduced significantly in the curcumin-treated group in comparison with the methotrexate-treated group. The ability of curcumin to inhibit methotrexate's cytotoxic effects was shown by the compound's ability to raise the mitotic index. According to this finding, curcumin approved to be a protective agent against genotoxic effects of methotrexate. Keywords: Curcumin, Chromosomal aberrations, Methotrexate, Mitotic index, Mice

Permeability Enhancement of Methotrexate Transdermal Gel using Eucalyptus oil, Peppermint Oil and Olive Oil(Conference Paper )#

Abstract

Objective: the idea of this study to improve transdermal permeability of Methotrexate using eucalyptus oil, olive oil and peppermint oil as enhancers.
Method: eucalyptus oil (2% and 4%), peppermint oil (2% and 4%) and olive oil (2% and 4%) all used as natural enhancers to develop transdermal permeability of Methotrexate via gel formulation. The gel was subjected to many physiochemical properties tests. In-vitro release and permeability studies for the drug were done by Franz cell diffusion across synthetic membrane, kinetic model was studied via korsmeyer- peppas equation.
Result: the results demonstrate that safe, nonirritant or cause necrosis to rats' skin and stable till 60 days gel was successfully formulated.<

Rosemary (Rosmarinus officinalis L.) is one of the most economically important species of the family Lamiaceae. Rosemary extract was examined by applying 2.2-diphenyl-1- picrylhydrazyl (DPPH) radical-scavenging assays. The result proved that rosemary extract had a higher antioxidant activity by absorption at a wavelength of 517a nm by using three different concentrations (0.5, 1.0 and 3) mg/ml which performed the absorbance at (2. 314, 0. 211 and 0.296) nm in comparison with control (21.8, 92.2 and 90) nm respectively. Extraction and estimation of curcumin compounds from turmeric (Curcuma longa) rhizomes by using different variables (solvent, time and temperature).

The study aimed to investigate the effect of different times as follows 0.5, 1.00, 2.00 and 3.00 hrs, type of solvent (acetone, methanol and ethanol) and temperature (~ 25 and 50)ºc on curcumin percentage yield from turmeric rhizomes. The results showed significant differences (p? 0.05) in all variables. The curcumin content which were determined spectrophotometrically ranged between (0.55-2.90) %. The maximum yield was obtained when temperature, time and solvent were 50ºC, 3 hrs and acetone, respectively.
