Optimum Conditions for Pyomelanin Production and Characterization from Local Isolates of Pseudomonas aeruginosa

Nowadays, there is increased interest in the biosynthesis of microbial melanin related to their numerous biological functions and applications in many fields, especially in medical fields, including immune-modulating, antimicrobial antibiotic, antiviral antivenin, anticancer, antitumor activity, and anti-biofilm activity. Pyomelanin is a hydrophobic macromolecule that is typically dark brown or black in color, formed by the oxidative polymerization of phenolic or indolic compounds. Pyomelanin is reported to be safe for consumption, thus providing a crucial strategy for biocontrol of biofilm. Furthermore, natural pyomelanin is known as a potent antioxidant, photoprotective, and free radical scavenging. Objective: This study focuses on the extraction and parametric optimization of melanin using Pseudomonas aeruginosa (P. aeruginosa) isolates. Materials and Methods: P. aeruginosa was isolated from various clinical sources, including urinary tract infections, burn patients, and wound infections. The isolate’s identification was done by biochemical, cultural, and VITEK-2 compact system. The isolates were subjected to pyomelanin production in nutrient agar supplemented with L-tyrosine, and the potential isolate was selected and employed for melanin production using nutrient broth supplemented with L-tyrosine. The pyomelanin-producing isolates were further identified through the PCR analysis technique. Different physical parameters were used for optimum pyomelanin yield. Results: The potential P. aeruginosa isolates were further identified using 16s rRNA. Pyomelanin production was better at 2.9 µg/mL at 100 rpm. The highest pyomelanin production was 3.07 µg/mL after 72 h incubation. The optimum pH at 7 was 3.12 µg/mL. The highest pyomelanin yield at 40°C was 3.03 µg/mL, while the maximum pyomelanin was produced using 2 g/L tyrosine at 2.99 µg/mL. UV-visible (UV-Vis) spectroscopy analysis for pyomelanin pigment was at 273.5 nm. The infrared spectrum exhibited a broad absorption band around 3280.82 cm−1 which corresponds to the presence of -OH and N-H groups. Conclusion: This study provides knowledge on factors affecting pyomelanin production using bacteria in broth media supplemented with L-tyrosine.

Production and Evaluation of Liquid Hydrocarbon Fuel from Thermal Pyrolysis of Virgin Polyethylene Plastics

   Pyrolysis of virgin polyethylene plastics was studied in order to produce hydrocarbon liquid fuel. The pyrolysis process carried out for low and high-density polyethylene plastics in open system batch reactor in temperature range of 370 to 450°C.

   Thermo-gravimetric analysis of the virgin plastics showed that the degradation ranges were between 326 and 495 °C. The results showed that the optimum temperature range of pyrolysis of polyethylene plastics that gives highest liquid yield (with specific gravity between 0.7844 and 0.7865) was 390 to 410 °C with reaction time of about 35 minutes. Abstract<p>The aim of this study was to identify the depth of the mouth and its shape in some local fish belonging to the Cyprinidae family, and the extent to which the depth of the mouth is related to the way of feeding and the nature of food as well as the feeding habits of those species collected specifically from the Tigris River, the results showed a relationship of depth oral cavity with head length was highly significant at (P < 0.01) for all studied species. Also, there was a highly significant relationship between the height of the pharyngeal tooth-bearing bone and the depth of the oral cavity for fish of this local family.</p>

Fortification of Meat Burger with Protein Isolate Extracted from Local Pumpkin Seeds

ABSTRACT: Protein isolate was achieved from local peeled non soaked pumpkins seeds by using petroleum ether with protein percentage of 53.15%. Protein isolate was used in manufacturing meat burger with two substitution10 and 20%. The shrinkage percentage for burger diameter was decreased from 25.5 to 16.6%, the sample with 10% substitution was distinguished in water holding capacity (WHC) which was 54.52%. Sensitive evaluation for these samples showed that the burger with 10% substitution was similar to the control.

Optimum diameter for laser beam and effect of temperature rise on the optical bistability hysteresis loops

 In this research, analytical study for simulating a Fabry-Perot bistable etalon (F-P cavity) filled with a dispersive optimized nonlinear optical material (Kerr type) such as semiconductors Indium Antimonide (InSb). An optimization procedure using reflective (~85%) InSb etalon (~50µm) thick is described. For this etalon with a (50 µm) spot diameter beam, the minimum switching power is (~0.078 mW) and switching time is (~150 ns), leading to a switching energy of (~11.77 pJ) for this device. Also, the main role played by the temperature to change the etalon characteristic from nonlinear to linear dynamics.

Prediction of Equilibrium Mixing Index and Optimum Mixing Time for Three solid materials in Fluidized Column

     Equilibrium and rate of mixing of free flowing solid materials are found using gas fluidized bed. The solid materials were sand (size 0.7 mm), sugar (size0.7 mm) and 15% cast iron used as a tracer. The fluidizing gas was air with velocity ranged from 0.45-0.65 m/s while the mixing time was up to 10 minutes.
