The quality of groundwater should be improved by keeping safe water sources from contaminants in protective way by doing regular measuring and checkup before it supplied for usage. Private Wells do not receive the same services that wells supplying the public do. Well owners are responsible for protecting their drinking water. This work was carried out in Badra city, Iraq from December 2017 to May 2018, six wells water were investigated to determine the general characteristics of wells as well as studying the effect of environmental factors on the quality of water. The study aims to detect the presence of carbapenems genes and the prevalence of antibiotic-resistant E. coli in the Tigris River. Samples were collected from three sites of the Tigris River: S1Adhamiya, S2 Medical city hospital, and S3 Abu Nuwas. It diagnosed 40 isolates of E. coli out of 67 isolates of bacteria by Vitek2. The antibiotic sensitivity was determined by the disk diffusion method. E.coli isolates were tested against 7 antibiotics these belonged to β-lactam, Carbapenem. In this study; a three-dimensional model was created to simulate groundwater in Al-Haydariyah area of the governorate of Al-Najaf. A solid model was created to utilize the cross sections of 25 boreholes in the research region, and it was made out of two layers: sand and clay. The steady-state calibration was employed in six observation wells to calibrate the model and establish the hydraulic conductivity, which was 17.49 m/d for sand and 1.042 m/d for clay, with a recharge rate of 0.00007 m/day.
