The isolates were screened according to their capability for pectinase production, screening process identified the best pectinolytic isolate and it was characterized by cultural and biochemical, as Pesudomonas sp. Pectinolytic enzyme producing bacterium Pesudomonas sp. was isolated from the Iraqi soil on nutrient agar plate. Optimiztion of process parameters were carried out by altering some of environmental conditions of chemo-physical environment for the production medium. The highest pectinase production was observed at 48 hrs of incubation at 35 °C with the initial pH of 6.0. Different nutrients and environmental conditions were investigated in terms of their effect on the production of extracellular pectinase using citrus pectin as substrate. The results exhibited that Yeast extract 0.15% with 0.5% of citrus pectin supported maximum pectinase production in the optimum conditions.
The inhibitory effect of Eucalyptus rostrata leaves extraction was investigated on multiple-antibiotic-resistant pathogenic bacteria (E.coli and S. aureus), isolated from Iraqi patients. The minimum inhibitory concentrations in a final concentration of 10 mg/ml. Tow fold dilutions was done from (12.5- 100) mg/ ml to examine the antibacterial effect of different concentrations of the plant extract on both bacteria. The study results revealed that Eucalyptus rostrata extract has a potential inhibitory effect on both gram negative and gram positive species. The current study supports the traditional approach of using Eucalyptus rostrata leaves extraction in treatment trails against bacterial infections.
Bacterial vaginosis (BV) is a common infection that occurs when the number of lactobacillus spp. bacteria (vaginal flora) decreases in the vaginal canal. The study aimed to determine the prevalence of Staphylococcus aureus within vaginosis in order to emphasize the importance of early detection and treatment. Totally, 90 vaginal swabs were collected using speculum and swabbing. The vaginal swabs were subjected to standard microbiological testing, which included microscopy, cultures (Blood agar and Mannitol salt agar), and antibiotic sensitivity testing. The results showed that out of 90 samples, only 40 S.aureus isolates were collected. S. aureus isolates showed maximum sensitivity to gentamic
... Show MoreA total of 172 clinical were obtained over 6 months. Klebsiella spp. was detected in 58 (33.7%) samples with a high percentage 29 (50%) in urine in female and low percentage 1(1.7%) in pus and burn swabs in male, and the vaginal swab was 1(1.7%). The female to male ratio was 3.1:1. PCR detection showed that 51(87.93%) out of 58 produce 108 bp. product with rpoB specific primer that represented K. pneumonia. Whereas 7(12.07%) showed PCR product with 343 bp by K. oxytoca specific primer (peh X), furthermore, the sequences of two selected isolates showed that the species related to K. oxytoca strain CAV1335, and to K. oxytoca strain CAV1374. Five selected isolates were re-tested by the gyr A primer, all were showed specific band product wit
... Show MoreFifty isolates of Bacillus sp. were subjected to the first and second screening to detect the ability to produce laccase enzyme and select the highest ones production of laccase on Petri plates containing nutrient agar supplemented with Cu2+.
Syringaldazine was used as an indicator and substrate for the determination of laccase activity. Three isolates, which consumed less time to developed pink color were tested for the production of laccase quantitatively. The effective isolate B16 with significant amounts of laccase 1.84 unit /ml was selected for laccase study.
The optimization studies revealed that the maximum laccase production was achieved when the production medium was at the following conditions: 5 days of incubation, tempe
The world is confronted with the twin crisis of fossil fuel depletion and environmental degradation caused by fossil fuel usage. Biodiesel produced from renewable feedstocks such as Jatropha seed oil or animal fats by transesterification offers a solution. Although biodiesel has been produced from various vegetable oils such as Jatropha seed oil, the reaction kinetics studies are very few in literature, hence the need for this study. Jatropha curcas seed oil was extracted and analyzed to determine its free fatty acid and fatty acid composition. The oil was transesterified with methanol at a molar ratio of methanol to oil 8:1, using 1% sodium hydroxide catalyst, at different temperature
... Show MoreAnimal fats are a good, promising and ethical alternative source for biodiesel production, but they need more complex treatments than vegetable oils. Iraqi butchery plants waste fats (sheep fat) which are suggested as feedstock to produce biodiesel. This type of fat contains a large quantity of free fatty acids (FFAs) (acid number 49.13 mg KOH/g of fat). The direct transesterification of such fats produce high amount of soap instead of desired biodiesel, so a pre-treatment step (to reduce FFAs) is necessary before transesterification. This step was done by esterification of the free fatty acids in the fat by adding ethanol and using 1% acid catalyst (H2SO4) for 30 minutes. The results showed that the acid number of sheep fat after pre-tr
... Show MoreNinety nine swabs were collected from patients with diabetic foot ulcers (DFU), all swabs were cultured on different selective media for screening, 46 isolates confirmed as S. aureus by API staph. The results of antibiotic susceptibility test revealed that all isolates were resistant to metronidazole, 34 isolates were resistant to cefoxitin, ceftriaxone, and meropenim, 23 isolates were resistant to ciprofloxacin and norfloxacin, 17 and 16 isolates were resistant to tetracycline and trimethoprim, respectively; while all isolates were sensitive to tigecycline. The results of minimum inhibitory concentration (MIC) that carried out by using vancomycin, tigecycline and linezolid for 8 isolates, MIC results were1-2 µg /ml
... Show MoreFour samples were collected from the wastewater of State Battery Manufacturing Company (SBMC); Babylon 2 factory in AL-Waziriya district, as triplicates. Physical and chemical measurements were carried out such as temperature, pH, Lead concentrations and their ranges were: (19.5-34.5) °C, (6.1-6.4) and (4.5-6.5) mg/L, respectively. Six dominant Bacillus spp. isolates were isolated from these samples; namely, Bacillus subtilis N1, Bacillus subtilis N2, Bacillus subtilis N3, Bacillus cereus N4, Bacillus cereus N5 , Bacillus cereus N6. These isolates were capable of removing Lead from aqueous solutions in a capacity reached 27.6 ± 1.4, 10.1 ± 1.7, 74.5 ± 0.7, 8.93 ± 2.8, 8.1 ± 3.5, 1.6± 0.7 mg/L, respectively. Whereas cell walls,
