Pseudomonas aeruginosa is considered as a developing opportunistic nosocomial pathogen and is well-known for its multidrug resistance that can be efficiently treated by a combination of antibiotics andefflux pump inhibitors (EPI). Therefore, the purpose of this study was to investigate the effect of curcumin as an EPI for the enhancement of the effectiveness of antibiotics against multidrug resistant (MDR) isolates ofP. aeruginosa. Susceptibility patterns of suspected bacteria was determined using the disc diffusion method andresistant bacteria were identified using chromogenic agar and 16S rDNA. The effectsof curcuminon the enhancement of antibiotics’s activity was evaluated usingthe broth microdilution method. The susceptibility patterns for 50 (67.6%) suspectedP. aeruginosaisolates showed that 36 (72%) of these isolateswere resistant to one of the used antibiotics, whereasonly 21 (42%) were MDR. The highest percentage of resistance was observedtoceftazidime (66%) followed by ciprofloxacin and levofloxacin (40%). Only 35 isolates were specified by chromogenic agar and 16S rDNAas P. aeruginosa. The minimal inhibitory concentration (MIC) of 35 isolates for ciprofloxacin resistant was between 4 and128 µg/ml while for ceftazidime was between 64and 512 µg/ml. After the addition of 50 μg/ml curcumin with ciprofloxacin, there wasa significant increase in the sensitivity (p≤ 0.01) of 13 MDR P.aeroginosa isolates whereas no differences in the sensitivity to ceftazidime were recorded before and after addition ofcurcumin. In conclusion, the results of this study show that curcumin can decrease the MIC value of ciprofloxacin in MDR isolates of P. aeruginosaand can be used as a native compound to enhance the treatment of resistant isolates with ciprofloxacin.
Over the past few decades, the health benefits are under threat as many commonly used antibiotics have become less and less effective against certain illnesses not only because many of them produce toxic reactions but also due to the emergence of drug-resistant bacteria. The clinical use of a combination of antibiotic therapy for Pseudomonas aeruginosa infections is probably more effective than monotherapy. The present study aims to estimate the antibacterial and antibiofilm activity of Conocarpus erectus leaves extracts against multi-drug resistant P. aeruginosa isolated from different hospitals in Baghdad city. One hundred fifty different clinical specimens were collected from patients from September 2021 to January 2022. All samples were

The values of minimum inhibitor concentration (MICs) for IMI and NIC (Separately) against P. aeruginosa were (16) ug/mL and (8) ug/ml respectively. Whereas, the concentration of NIC with IMI (as combined) for biofilm inhibition was 1 ug/ml for NIC and 4 ug/ml for IMI. The combining of NIC with IMI showed synergistic efficacy against formation of bacterial biofilm (at MIC levels). These results provide a conclusion that NIC combined with IMI is can be considered as a successful prospective treatment against the biofilm pr

Aim: The current study aimed to make an aqueous crude extract from the soil Proteus mirabilis isolate with the use of the gas chromatography-mass spectrometry (GC-MS) technique for its analysis, and then, study the impact of the extract on clinical isolates of Pseudomonas aeruginosa. Methods: Preparation of crude extracts from P. mirabilis (both organic and aqueous), which were then analyzed by GC-MS to detect the bioactive ingredients. Furthermore, the extract’s capability to i

This organism appears to improve resistance to many antimicrobial agents and a high percentage of clinical isolates of P. aeruginosa exhibit multidrug resistance (MDR) phenotype . The purpose of this study is to screen the antibiotic susceptibility patterns and the prevalence of qacE delta1 gene among bacterial isolates. Accordingly, 145 samples were collected from different clinical sources from patients who admitted to different hospitals in Baghdad city in a period ranged 23/8/2018-1/1/2019. The isolates were diagnosed as P. aeruginosa based on routine b

The test organisms include 3 Gram negative bacteria such as Echerichia coli, Klebsiella pneumoniae, Salmonella typhi and 2 Gram positive bacteria such as Staphylococcus aureus and Staphylococcus epidermidis. The algal cell mass was extracted in 90% Methanol and 90% Ethanol and further concentrations of 0.5, 1, 3, 5, 10, 20 mg/ml were made for each extract. Antagonistic effect was tested by using agar well-diffusion method. Methanolic crude extract showed strong antibacterial activity against all tested bacteria, while ethanolic crude extract showed moderate activity. These findings suggest the possi

In this study we evaluated the synergistic effect of S. striata hydroalcoholic extract (SSE) and commercially available antibiotics against P. aeroginosa and Methicillin- resistant Staphylococcus aureus (MRSA). The resazurin-based microdilution method was used to determine the minimum inhibitory concentration (MIC) values of plan extract and standard antibiotics. The interaction between standard antibiotics and SSE was evaluated by using checkerboard method. The results of this study revealed that SSE enhance the antibacterial activity of antibiotics. Pseudomonas aeruginosa P1 was selected as the highest protease producer, which next identified as P. aeruginosa. It was found that the optimum culture conditions for protease production in submerged culture was in the tryptic - soya broth medium at 37° C with pH 8 for 48 hours. In addition, the study i

P. aeruginosa has a well-known biofilm forming ability. The expression of polysaccharide encoding locus (pelA gene) by P. aeruginosa is essential for this ability. The purpose of the current research was to determine the biofilm formation in P. aeruginosa isolated from clinical samples and to evaluate the role of the selected PelA gene in biofilm formation using PCR method in Iraqi patients. Results revealed that 24 (96%) isolates were found to have the ability to form biofilm that was remarkably related to gentamicin resistance. Moreover, the pelA gene was found in all biofilm-producers. In c

Out of 143 patients suffering from burns, urinary tract infections (UTI), respiratory tract infections and cystic fibrosis obtained from previous study by Mahmood (2015), twenty two isolates (15.38%) were identified from (11) hospitals in Iraq, splitted into three provinces, Baghdad, Al-Anbar and Karbala for the duration of June 2017 to April 2018. Characterization was carried out by using microscopical, morphological and biochemical methods which showed that all these isolates belong to P. aeruginosa.
