A new ligand 3-hydroxy-2-(3-(4-nitrobenzoyl) thiouriedo) propanoic acid (NTP) where synthesized by reaction of 4-nitro benzoyl isothiocyanate with serine amino acid. The ligand was characterized by FT-IR, NMR spectra and the elemental analysis.    In the present study, serum GOT(Glutamic Oxalacetic Transaminase) was purified, the purified enzyme showed the maximum activity at 37ºC and pH 7.5. During purification of serum GOT ion exchange chromatography lead to four separate forms (termed I, II, III and IV). The bottom wall of the enclosure is maintained at higher temperature than that of the top wall which keeps in oscillation motion, whereas the left and right walls are well insulated. The differential temperature of the bottom and top walls changed several times in order to accurately characterize the temperature distribution over a considerable range of Richardson number.
