The current study aimed to detect the effect of gentamicin stress on the expression of hla (encodes hemolysin) and nuc (encodes nuclease) genes of Staphylococcus aureus. Fifty-eight isolates identified as S. aureus were isolated locally from different clinical specimens. Disk diffusion method was used to detect the resistance to S. aureus. The minimum inhibitory concentration (MIC) of gentamicin was estimated by broth microdilution method. hla and nuc genes were determined by polymerase chain reaction technique. The biofilm was evaluated using the microtiter plate method in the presence and absence of gentamicin at sub-MIC. The results showed that 18 (31%) and 40 (69%) S. aureus isolates were sensitive and resistant to gentamicin, respectively. All S. aureus isolates succeeded in forming biofilm. However, eight (13.79%), 28 (48.28%) and 22 (37.93%) isolates produced weak, moderate and strong biofilms respectively. In most isolates, gentamicin at sub-MIC decreased biofilm intensity. Due to gentamicin stress, the hla and nuc genes were upregulated in S. aureus biofilm.
This study is an investigation of the drugs effect on some pathogenic Acanthamoeba isolated from Iraqi waters, where the problem of environmental adaptation that characterizes this organism in addition to being a reservoir for many pathogenic microorganisms that take shelter in it to escape disinfectants and medicines is sometimes difficult to treat it with traditional treatments. Twenty water samples were collected from different water regions in Iraq, namely the Dokan Lake, Tigris River, Euphrates River and Najaf Sea, 5 samples from each source. Acanthamoeba was isolated from water samples on NNA and PYG media, using an inverted microscope with an electron microscope to determine their phenotypic features. PCR and
... Show MoreThe pathogenicity of S. saprophyticus was studied in mice. A group of white mice were injected transurethrally using a catheter with S. saprophyticus S67 cell suspension in a concentration reached 109 CFU/ml. concomitantly, the role of its peptidoglycan in the pathogenicity was studied by injecting another group of mice with 0.3 mg/0.2 ml of partially purified S. saprophyticus S67 peptidoglycan extract. After autopsy, kidneys and urinary bladder showed several histopathological changes both in cells and peptidoglycan injected mice, included: hydropic degeneration, glomerulus shrinkage, congestion of renal vessels, infiltration of inflammatory cells, and dekeratinization in urinary bladder.
Background: The etiology of ischemic heart disease (IHD) is believed to have an immunological component. Association with human leukocyte antigens (HLAs) has been previously reported, particularly with DR6.
Patients and methods: 75 cardiac patients were admitted to the coronary care unit, Baghdad Teaching Hospital over the period October 2008-May 2009 with the clinical diagnosis of acute
coronary syndrome and STEMI myocardial infarction their ages range was (25-82) years the number of male was (55) (73.3%) and female was (20) (26.7%). All cases have routine ECG, cardiac marker’s measurements, routine haematological, Biochemical test and 2mls of blood reserved for HLA study.
Results: It was found that H
The present study aimed at shed light on the association between HLA-class I antigens (A, B and Cw) and brain tumours (meningioma and glioma) in the basis of their individual frequencies or two-locus association A total of 52 brain tumour patients were enrolled in this study, with an age range of 7-68 years. The patients were divided into two clinical groups; meningioma (20 cases) and glioma (22 cases), while the remaining 10 cases represented other types of brain tumour. Control samples included 47 Iraqi Arab apparently healthy blood volunteers, with an age range of 15-50 year. Three HLA antigens showed a significant increased frequency in total patients as compared to controls. They were B13 (34.6 vs. 6.5%), B40 (15.4 vs. 2.2%) and Cw3
... Show MoreKlebsiella pneumoniae capsular polysaccharide (CPS) antigen was evaluated for their capability to increase immune responses. And, CPS neutralizing antibodies were approved as the main response to vaccination in many disease. Therefore, killed Stapthylococcus aureus bacteria was employed to evaluate K. pneumoniae CPS adjuvanticity. The mice groups were immunized (orally, intra-peritoneally and by swab skin)with a dose of (25μl of formalin killed S. aureus (1.5 x 108) with a CPS at dose 175μl/kg at a conc.50 μg/ml) vaccination occurred in first day then recurrent vaccination as booster dose beyond seven days. After first 7 days, the results revealed elevation of IL2,4,10,12 and IgG levels occurred mainly in oral and swab skin groups, an

Show MoreFifty one patients with serologically confirmed brucellosis and 70 healthy controls were phenotyped for HLA-A, -B, -DR and -DQ antigens by using standard microlympho-cytotoxicity method, and lymphocytes defined by their CD markers (CD3, CD4, CD8 and CD19). The results revealed a significant (Pc = 0.001) increased frequency of HLA-DR8 (41.18 vs. 10.0%) in the patients . A significant increased percentage of CD8+ lymphocytes was also increased in the patients (25.15 vs. 22.0%; P = 0.006), while CD3+ lymphocytes were significantly decreased (75.1 vs. 79.4%; P = 0.02).
Background: Human leukocyte antigen (HLA) is the most polymorphic genetic system in main.
Background: Human leukocyte antigen (HLA) is the most polymorphic genetic system in man. The genes of this region seem to influence susceptibility to certain diseases.
Patients and methods: Polymerase chain reaction-Sequence Specific Primers PCR-SSP is the method used to asses HLA-typing of 100 blood samples of 60 AIH patients and 40 healthy normal controls.
Results: An increased frequency of HLA-DR3, DR4 and DR7 was observed for patients group versus control group with P-value (0.0001, 0.05, and 0.001) respectively, while DR*0211 (DR2) may be formed the basis for protection against the disease. HLA-DQ on the other hand, yielded on association in Iraqi patients with AIH.

Reactive arthritis (ReA) has been as joint developing after infection, it belongs to spongylo arthritis (SpA). The etiology of this disease was multi factorial, the combination between genetic and environmental factors for triggering this disease. This study included 75 Iraqi Arab patients and 39 healthy control. Urine samples and blood were collected from each subject. The results showed that Escherichia coli bacteria (E. coli) was isolated from 32% of urine samples. HLA-B*27 allele frequencies was higher in ReA patients infected with E. coli. This lead to suggest that E. coli may be trigger factor in ReA patients with UTI which had HLA-B*27 positive.
