The aim of this study was the discrimination of Salmonella isolated from chicken and their feed and drinking water for the epidemiological control of salmonellosis. Totally, 289 samples, including 217 chicken cloaca swabs, 46 water, and 26 feed samples were collected from five different farms in Karbala governorate, Iraq. Conventional bacteriology tests, API 20E, Vitek 2, and serology were used for bacterial identification. Random amplified polymorphic DNA (RAPD)-polymerase chain reaction (PCR) was applied to analyze the genetic relationships among Salmonella isolates. The isolation rate of Salmonella spp. was 21.1% (61/289). While the water samples constituted the highest rate (30.4%), a rate of 21.7% was reported for the cloaca swabs, with no isolate at all from chicken feed. Vitek 2 was able to identify some isolates to the serotype level, such as S. Enteritidis, S. Paratyphi B, and S. Paratyphi C. However, the isolates were diagnosed as S. enterica by API 20E, and as S. enterica subsp. arizonae through serology. Analyzing the samples by the RAPD-PCR assay showed the presence of genetically different Salmonella spp. Dendrograms created by the GelJ software successfully delineated the genetic relationships. Therefore, RAPD-PCR can be used as a surrogate tool for the fast, reliable, and accurate detection of Salmonella in epidemiological surveys when compared with other biochemical-based identification methods.
study was conducted on a stretch of Tigris river crossing Baghdad city to determine the concentration of some chlorophenols pollutants. Aqueous samples were preliminary enriched about 500 times and the chlorophenols have determined using high performance liquid chromatography HPLC. Limits of detection LOD were (0.007–0.012 mg L-1), relative standard deviations RSD% were 2.4%–5.59% and relative recoveries were 51.06%– 104.07%. The existence of chlorophenols in Tigris river was in the range 0.023–4.596 mg L-1. The developed method suggested in this study can be applied for routine analysis and monitoring of chlorinated phenols in environmental aqueous samples.
Leuconostoc bacteria was isolated from local pickled cabbage (Brassica oleracea capitata) and identified as Leuconostoc mesenteroides by morphology,biochemical and physiological. The local isolated L. mesenteroides bacteria under the optimal conditions of dextran production showed that, the highly production of dextran was 7.7g achieved by using a modified natural media comprised of 100ml whey, 10g refined sugar, 0.5g heated yeast extract, 0.01g CaCl2, 0.001g MgSO4, 0.001g MnCl2 and 0.001g NaCl at pH 6 and 25̊C for 24 hr of fermentation and by using 1ᵡ106 cell/ml as initial inoculums volume. Salmonella typhimurium ] were obtained from the Central Health Laboratory . Both sensitivity tests (MIC , MBC and wells method ) against these bacteria were performed by using the aqueous of leaves extract of Marjoram plant. The results cleared that the values of MIC for Vibrio cholera serotypes Ogawa and Inaba were 100 mg/ml , while the value of MBC was 200 mg/ml. The value of the Inhibition zone at 100 mg /ml concentration for both Ogawa and Inaba were 13 mm and 9 mm respectively. Our results showed that the three types of Salmonella didn’t show any inhibition zone at 200 mg/ml .
