PURIFICATION AND CHARACTERIZATION OF AMYLASE EXTRACTED FROM LOCAL WHEAT PLANT

This study was aimed to test four types of different plants for the purpose of selecting the optimal plant as a source of the amylase enzyme, including wheat, barley, rice, and potatoes. Among these plants, wheat was selected as the best plant source for enzyme extraction because it possessed the highest effectiveness of the enzyme specific activity (3.0 U/mg protein). Furthermore, sodium acetate buffer (0.2 M, pH 6.0) was determined to be the optimal extraction buffer with a 1:5 (w:v) ratio after 75 min, and it provided 68 U/mg protein. The results revealed that the molecular weight was 180 kDa by gel filtration and Native-PAGE. The optimal pH and temperature for enzyme activity were 7 and 40 °C respectively. The enzyme activity was stable against pH values range of 6-8. The enzyme retained 79% of its total activity after incubation for one hour at 50 °C and retained 7% (lost 93%) of its activity at 70 °C. EFFECT OF SUPPLEMENTATION FENUGREEK OIL TO THE DIET ON THE PHYSIOLOGICAL, ANATOMICAL AND HISTOLOGICAL TRAITS OF BROILERS

This study was aimed to evaluate the effect of supplementing fenugreek seed oil to the diet on the physiological, anatomical and histological characteristics of broilers. A total of 200 chicks one day-old of (Ross308) were used in this study and divided into 4 treatments, 5 replicate for each treatment by 10 chicks in each replicate. The treatments were T1(control diet) = without additives (basal diet), T2 = basal diet + 0.25 ml fenugreek oil /  kg, T3 = basal diet + 0.50 ml fenugreek oil /  kg and T4 = basal diet + 0.75 ml fenugreek oil /kg. This study was aimed to estimate the antioxidants, antibacterial, and antifungal activities of methanolic extract from soybean seeds against pathogenic microorganisms. The methanolic extract was prepared from defatted soybean flour. Total phenolics content (TPC), total flavonoid content (TFC), and the major phenolic, flavonoid, and isoflavone compounds in soybean extract were estimated by HPLC.
