The production of reactive oxygen species in most diseases including osteoarthritis (OA) is confirmed to be the most destractive process to human cells.  To investigate the role of free radicals in patients with OA and by comparing lipid peroxidation marker maloan dialdehyde (MDA) and Vit E as one of the potent antioxidant in Sera of (OA) patients with those in healthy population.  The study includes 34 patients with knee OA diagnosed by doctors using different diagnostic parameters techniques , 18 females and 16 males and their age ranged 45 – 78 years , in addition to  32 healthy control 16 males and 16 females. Aspartate aminotransferase was purified from urine and serum of patients with type 2 diabetes in a 2 steps procedure involving dialysis bag and sephadex G-25 gel filtration (column chromatography). The enzyme was purified 346.23 fold with 1467% yield and 3.46 fold with 142.85% yield in urine and serum of patients with type 2 diabetes respectively. The purified enzyme showed single peak. The results of this study revealed that AST activity of type 2 diabetes urine and serum increased significantly (p<0.001) compared with control group.
