Reduction of cell viability and alteration of genomic DNA methylation by phthalates in a normal human fibroblast (NHF) cell line in vitro

Phthalates are ubiquitous environmental contaminants widely used as plasticizers in poly vinyl chloride products, and growing evidence implicates them in metabolic activity reduction and epigenetic dysregulation. However, their direct effects on non-transformed human cells remain incompletely characterized. The present in vitro study evaluated the reduction in cell viability and DNA methylation–modulating effects of three widely distributed phthalates, di(2-ethylhexyl) phthalate (DEHP), di-n-butyl phthalate (DnBP), and di-isononyl phthalate (DINP), on normal human fibroblast (NHF) cells. Cells were exposed to serial concentrations (31.25–1000 μg/mL) for 72 h, and the viability was assessed by MTT assay, followed by IC50 determination via nonlinear regression. Global DNA methylation was subsequently quantified at the IC50 concentration by ELISA-based measurement of 5-methylcytosine (5-mC). All three phthalates produced significant concentration-dependent reductions in cell viability. Constraining the regression plateaus to 100% and 0% viability yielded estimated IC50 values of 19.2 μg/mL for DEHP, 43.4 μg/mL for DnBP, and 13.1 μg/mL for DINP; as viability fell below 50% at the lowest concentration tested, these values represent extrapolation below the sampled range, their confidence intervals overlap, and no potency ranking is asserted. At fixed high-effect concentrations, DEHP and DINP significantly elevated global 5-mC levels to 189.18% and 159.56% of control, respectively, whereas DnBP induced only a non-significant increase (120.06%). These findings indicate that phthalates compromise both the viability and epigenetic integrity of NHF cells in a congener-specific manner, underscoring their potential contribution to methylation-associated human pathologies.

Esterification of Free Fatty Acid with High Chain Alcohol for Biodiesel Production Using Semi-Batch Reactive Distillation

The esterification of oleic acid with 2-ethylhexanol in presence of sulfuric acid  as homogeneous catalyst was investigated in this work to produce 2-ethylhexyl oleate (biodiesel) by using semi batch reactive distillation. The effect of reaction temperature (100 to 130°C), 2-ethylhexanol:oleic acid molar ratio (1:1 to 1:3) and catalysts concentration (0.2 to 1wt%) were studied. Higher conversion of 97% was achieved with operating conditions of reaction temperature of 130°C, molar ratio of free fatty acid to alcohol of 1:2 and catalyst concentration of 1wt%. A simulation was adopted from basic principles of the reactive distillation using MATLAB to describe the process. Good agreement was achieved.

   The aim of this paper is to present the first record of ctenophore species Pleurobrachia pileus (O. F. Müller, 1776) in the coral reef as was recently found in Iraqi marine waters. The specimens were collected from two sites, the first was in Khor Abdullah during May 2015, and the second site was located in the pelagic water of the coral reef area, near the Al-Basrah deep sea crude oil marine loading terminal. Three samples were collected at this site during May 2015, February and March 2018 which showed that P. pileus were present at a densities of 3.0, 2.2 and 0.55 ind./ m3 respectively. The species can affect on the abundance of other zooplankton community through predation.

This work presents the fabrication and the characterization of a relative humidity sensor based on a polymer-coated photonic crystal fiber that operates in a Mach- Zehnder Interferometer (MZI) transmission mode. The fabrication of the sensor involved splicing a short (1 cm) length of Photonic Crystal Fiber (PCF) between two single-mode fibers (SMF). It was then coated with a layer of agarose solution. Generation of True Random TTL Signals for Quantum Key-Distribution Systems Based on True Random Binary Sequences

A true random TTL pulse generator was implemented and investigated for quantum key distribution systems. The random TTL signals are generated by low cost components available in the local markets. The TTL signals are obtained by using true random binary sequences based on registering photon arrival time difference registered in coincidence windows between two single – photon detectors.
