Expression of virulence and antimicrobial resistance genes among Escherichia coli clinical isolates from blood and stool samples
...Show More Authors
Objective:

As major nosocomial pathogens, Escherichia coli isolates exhibit antibiotic resistance and also express adhesive structures and antibiotic resistance genes. The objective of this study was the comparison of virulence gene expression of extended-spectrum beta-lactamase (ESBL)-producing E. coli between blood and stool samples.

Methods:

In this study, 20 E. coli clinical isolates (10 ESBL-producers including 5 from blood, 5 from stool samples and 10 non-ESBL-producer strains) were included. The existence of fimA, kpsMII and cdt (adhesives and toxin), acr-ab (efflux-encoding) and bla CTX-M1 genes were confirmed by PCR. The quantitative real-time PCR was performed for evaluation of gene expression.

Results:

ESBL-producing E. coli isolates from stool samples could express fimA, kpsMII and cdt genes significantly higher than blood samples, whereas those isolates from blood samples significantly expressed the acr-ab (efflux-encoding) genes. In addition, the bla CTXM1 gene was expressed among isolates from stool samples significantly higher (P = 0.022) than those from blood samples according to the analysis of variance (ANOVA) test. In addition, among non-ESBL-producers, the expression of fimA, kpsMII and cdt genes was significantly lower than ESBL-producing isolates from blood samples, but not significantly different than those from stool samples. Moreover, the expression of acr-ab genes was significantly lower than those from stool samples.

Conclusion:

The results exhibited that the expression of virulence genes among clinical isolates of E. coli is not the same or similar in various conditions or from various clinical origins. Thus determining the profile of gene expression in each of clinical situations can be helpful in tracking the infectious pathogens. ESBL-producing strains possibly have regulatory factors for inducing higher virulence gene expression.

Synthesis of New Nucleoside Analogues From Benzimidazole and Evaluation of Their Antimicrobial Activity
...Show More Authors

Our goal in this research, some new nucleoside analogues was synthesized. Starting from ?-D glucose which was converted to per acetylated ?-D gluco pyronoside then converted to active from(1-Bromo Sugar (2) as a sugar moiety. The base moiety 2-substituted benzimidazole was prepared from condensation of phenylene diamine with different aromatic aldehydes, which were subjected to amino alkylation via Mannich reaction forming new nucleobase derivatives. Condensation of nucleobase with bromo sugar through nucleophilic substitution of anomeric carbon with nitrogen forming new protected nucleoside analogues then hydrolyzed with sodium methoxide in methanol to obtain our target, the free nucleoside analogues. All prepared compound were identified b

Production of fibrinolytic protease from various fungal isolates and species 2.Determination of optimum conditions for enzyme production from Pleurotus ostreatus
...Show More Authors

The optimum conditions for production of fibrinolytic protease from an edible mushroom Pleurotus ostreatus grown on the solid medium , Sus medium, composed of Sus wastes (produced from extracted medicinal plant Glycyrrhiza glabra) were determined. Addition of 5% of Soya bean seeds meal in Sus medium recorded a maximum fibrinolytic protease activity resulting in 7.7 units / ml. The optimum moisture content of Sus medium supplemented with 5% Soya bean seeds meal was 60% resulting in 7.2 units / ml.Pleurotus ostreatus produced a maximum fibrinolytic protease activity when the spawn rate,pH of medium and incubation temperature were 2,6 and 30°C, respectively. The maximum fibrinolytic protease activity was 7.6 units / ml when incubat

Green Biosynthesis of Silver Nanoparticles from Moringa oleifera Leaves and Its Antimicrobial and Cytotoxicity Activities
...Show More Authors

The plant occupied the largest area in the biosynthesis of silver nanoparticles, especially the medicinal plants, and it has shown great potential in biotechnology applications. In this study, green synthesis of silver nanoparticles from Moringa oleifera leaves extract and its antifungal and antitumor activities were investigated. The formation of silver nanoparticles was observed after 1 hour of preparation color changing. The ultraviolet and visible spectrum, Fourier transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, and transmission electron microscopy techniques were used to characterize synthesis particles. Ultraviolet and visible spectroscopy showed a silver surface plasmon resonance band at 434

Molecular Alterations in IDH 1/2 Genes among Iraqi Adult Acute Myeloid Leukemia Patients
...Show More Authors
BACKGROUND:

The recurrent somatic variations in IDH1/2 genes in AML play imperative roles in epigenetic dysregulation and the pathogenesis of AML, which could be useful prognostic markers for risk stratification.

AIM:

The aim of the study was to detect the frequency of R132 mutations in the IDH1 gene and R140Q mutation in the IDH2 gene with their treatment outcomes.

Effect of laparoscopic sleeve gastrectomy on blood sugar and blood pressure
...Show More Authors

Background: Surgery is one and may be the most effective method to treat obesity. In the last decade, Laparoscopic Sleeve Gastrectomy is perceived to be less invasive, technically simple, less morbid and more popular form of bariatric surgery.

Objectives: This study aims to assess the effect of Laparoscopic Sleeve Gastrectomy on Fasting Blood Glucose Levels and Blood Pressure.

Methods: A prospective controlled study in which 50 obese patients were involved, 36 of patients have hypertension and type 2 diabetes mellitus , 7 patients have type 2 diabetes mellitus only, and 7 patients don’t have hypertension or type 2 diabetes. All patients were submitted to Laparosco

Detection of biofilm formation and antibiotics resistance of Staphylococcus spp. isolated from humans’ and birds’ oral cavities
...Show More Authors

Background: Staphylococcus spp. are widely distributed in nature and can cause nosocomial, skin infections, and foodborne illness, and it may lead to severe financial losses in birds by causing systemic infection in numerous organs. Aim: This study was conducted to determine the prevalence of Staphylococcus spp. in humans and birds in Baghdad city. Methods: Seventy-six oral cavity swabs were collected, including 41 from birds and 35 from breeders. All samples were examined by bacteriological methods and identified by using the VITEK technique, the samples were then further studied to test the ability of biofilm formation, and MDR factors and MAR index were tested with the use of seven antibiotics. Results: Among the 76 oral swa

Introduction: Trichophyton indotineae has emerged globally as a highly contagious, terbinafine-resistant dermatophyte causing treatment-refractory superficial mycoses, posing a significant public health threat. This study evaluated the epidemiology of dermatophytosis in Babylon Province, Iraq, and performed molecular identification and phylogenetic analysis of local clinical isolates. Methods: Clinical specimens from 258 patients aged 1 to 71 years were analyzed. Following phenotypic characterization, genomic DNA was extracted from three representative isolates. Polymerase chain reaction amplified the internal transcribed spacer region, followed by DNA sequencing and neighbor-joining phylogenetic analysis. Results: Tinea corporis was
