MRSA is one of the major pathogens in hospitals and the community, which have the ability to produce biofilm as a virulence factor, the impact of chalcone on biofilm formation, the synergism effect of chalcone and antibiotic in both in vitro and in vivo experiments, the gene expression of virulence genes (srtA, fnbA, fnbB) before and after treatment of it on MRSA biofilm cells in vitro, all these were the prime aims of this study. Chalcone at MBIC (20 μg/ml), significantly reduced the biofilm formation to 21.45% and at sub MBIC (15 μg/ml) to 36.58 %. While, Chalcone at MIC(5 μg/ml) reduced MRSA planktonic cells to 49.61%. Susceptibility of MRSA isolates against eight antibiotics showed that all isolates were sensitive to vancomycin and none of the isolates developed susceptibility to erythromycin. The combinatorial effect of chalcone at 5 μg/ml and vancomycin at MIC of (1 μg/ml) on MRSA planktonic cells was reduced it from 70 to 23.3% , and in combination with erythromycin at 32 μg/ml, was decreased from 53.1% to 22% and the effect of chalcone at sub MBIC (15 μg/ml) when combined with vancomycin was reducing the biofilm formation from 87% to 27.6 and with erythromycin from 55.1% to 23.8%. Combinatorial phenotypic effect of the antibiotics and chalcone (at sub MBIC), in vitro came in line with the result of in vivo experiment and the results showed decrease in the expression of fnbA, fnbB and srtA genes in tested isolates in the presence of chalcone at sub MBIC. In our study, we demonstrated that chalcone exhibited significant effect in biofilm formation of MRSA strains, which can be considered as promising antimicrobial agents that can be used for prevention of MRSA adherence or as adjunct to antibiotics in conventional therapy.
The pathogenicity resulting from Staphylococcus aureus infection has remarkable importance as one of the community-associated bacterial infections, due to the virulent ability of these bacteria to produce biofilms. This study was designed to detect biofilm production in clinical isolates from samples of wounds and urinary tract infections. The expression levels of the icaA gene that is responsible of slime layer production in biofilms was compared in isolates with different biofilm producing capabilities. Fifty seven samples that included 32 samples from urine and 25 samples from wounds were collected from Alwasti Hospital, Al-Kindi Teaching Hospital, and Alzahraa Clinic, Baghdad, Iraq. The bacteria was identified accor

Show MoreObjectives: The current work aimed to reveal the impact of gentamicin on the fibronectin binding proteins (fnbp) gene expression and its relation to biofilm and agr type in Staphylococcus aureus. Materials and Methods: A total of 25 S. aureus isolates were enrolled in this study previously isolated from different specimens. Identification confirmation and methicillin resistance were achieved by amplification of 16SrRNA and mecA. Multiplex polymerase chain reaction (PCR) based assay was employed to evaluate the agr typing. The gene expression of fnbA and fnbB genes was tested by real-time PCR technique. Minimum inhibitory concentration was estimated by micro broth dilution methodology. Microtiter plate method was performed to determine the a

Among 8 isolates, 5 (62.5%) S. aureus isolates were found to be enterotoxigenic, most of isolates produced at least two types of Staphylococcal enterotoxins (SEs). The production of enterotoxins in the presence or absence of Thymol extracts (aqueous and alcoholic) were estimated using a reversed passive latex agglutination (SET-RPLA) kit. The extracts reduced enterotoxin production compared with the control. Enterotoxin inhibition was observed for enterotoxin C production at minimal inhibitory concentrations (MIC) at 400 µg/ml, whereas production of enterotoxins A, B, and

To elucidate the anti- Methicillin resistance Staphylococcus aureus (MRSA) effect of pomegranate alone and in combination with moxifloxacin fluoroquinolone. A total of five clinical isolates of MRSA (ATCC 43300) were used in the study. Disc diffusion method was used to determine the anti-MRSA effect of pomegranate and/or moxifloxacin by using Mueller-Hinton agar. Minimal inhibitory concentration (MIC), fractional inhibitory concentration (FIC) of moxifloxacin and pomegranate were calculated, the dynamic picture of the bactericidal effect of pomegranate and/or moxifloxacin was determined. SPSS version 20.00 was used for data analysis. Zone of inhibition (ZOI) of moxifloxacin was 19.67±4.84mm which was not significant compared with pomegrana
... Show MoreStaphylococcus aureus, which includes the methicillin-resistant S. aureus (MRSA), is a significant human pathogen producing different toxins and results in many different infection types, which include bacteremia, soft-tissue infections, as well as staphylococcal food poisoning. S. aureus is an important food-borne pathogen of humans due to ingestion of food containing enterotoxigenic strains. Detecting S. aureus femA and mecA genes was evaluated with the use of a Loop-mediated Isothermal Amplification Method (LAMP). The accuracy of this approach was similar to that attained using the approach of the conventional polymerase chain (PCR). Those two methods characterized 43 isolates of MRSA which

S. aureus is considered as one of the most important pathogenic bacteria, having the ability to activate certain genes that encode for heat stable enterotoxins and cause Staphylococcal food poisoning. Thus, this study aimed to determine the prevalence of multi resistant Staphylococcus aureus that produce enterotoxins in different sources of food . Forty nine isolates were identified as S.aureus, according to morphological and biochemical tests. They were isolated from 387 different food samples from several randomly covered restaurants
