Optimization of ultrasonic-assisted extraction and in vitro determination of the cytotoxic effect of ethyl acetate fraction of wild Amaranthus viridis on SKGT-4, AGS and A431 cell lines

Amaranthus viridis L. belongs to the Amaranthaceae family. It is a rich source of numerous phytochemicals and amino acids. The objective of this work was to optimize Ultrasound-Assisted Extraction (UAE) based on the extraction yield and Thin-Layer Chromatography (TLC) profile under different conditions, to compare the optimized UAE to the Soxhlet extraction method and evaluate the cytotoxic effects of the ethyl acetate fraction of the 80 % ethanolic extract on the SKGT-4 (human esophageal adenocarcinoma), AGS (human gastric adenocarcinoma) and A431 (human epidermoid carcinoma). A one-factor at a time experiment was carefully designed to assess the influence of the following factors on the extraction: time, frequency, solid-to-solvent ratio and aqueous ethanol concentration. Soxhlet extraction using 80 % aqueous ethanol was done for defatted plant material, then fractionation using chloroform, ethyl acetate and n-butanol. Cytotoxicity of ethyl acetate fraction was evaluated using the MTT assay on AGS, A431 and SKGT-4 cell lines. The results indicated that in the UAE, the solid-to-solvent ratio has the most significant effect on yield. Soxhlet extraction proved to be more efficient than UAE in terms of TLC profiles. The cytotoxicity of the ethyl acetate fraction exhibited cytotoxic activity against the tested cell lines in a concentration-dependent manner. Thus, selecting a particular extraction method depends on the target compounds. Soxhlet is preferred for gaining certain compounds that require heat for their extraction. The ethyl acetate fraction showed a cytotoxic effect on various cell lines related to cell components and their interactions with phytochemicals present in this fraction.

This current study investigated the effects of two newly synthesized Quercetin Schiff bases containing 2-amino thiadiazole-5-thiol (Q1), and its benzyl derivatives (Q2) on MCF-7 human breast cancer cells. This effect was studied by adding increasing concentrations  of  Terpinen-4-ol to  culture wells containing mouse macrophages that were previously incubated with the promastigote forms of the parasites for 24 hours .Then, they were incubated for another 24 hours with increasing concentrations of Terpinen-4-ol.  After, Parasites were enumerated into macrophages in wells either treated with Terpinen-4-ol or in control wells.

The study effect of weight fraction on thermal and electrical conductivity for unsaturated polyester composite alone and hybrid

In this research prepared two composite materials , the first prepared from unsaturated polyester resin (UP) , which is a matrix , and aluminum oxide (Al2O3) , and the second prepared from unsaturated polyester resin and aluminum oxide and copper oxide (CuO) , the two composites materials (Alone and Hybrid) of percentage weight (5,10,15)% . All samples were prepared by hand layup process, and study the electrical and thermal conductivity. The results showed decrease electrical conductivity from (10 - 2.39) ×10-15 for (Up+ Al2O3) and from (10 - 2.06)×10-15 for (Up+ Al2O3+ CuO) .But increase thermal conductivity from( 0.17 - 0.505) for (Up+ Al2O3) and from (0.17 - 0.489) for (Up+ Al2O3+ CuO).

The present study aimed to evaluate the anti-inflammatory effects of two different extracts on chronic inflammation induced by cotton pellets in rats. Methods: Loranthus seed was extracted by maceration with absolute methanol, in which dry Loranthus seeds were triturated in a mortar and macerated with 500 mL of methanol. After 24 h, the mixture was filtered, and the residue was re-extracted. The filtrates were combined and dried under vacuum. The mixture was mixed with 100 mL of distilled water and fractionated by petroleum ether and ethyl acetate using 70 mL × 3 times each. The organic fractions were dried, filtered, and evaporated to dryness.

Cytotoxicity of purified methionine γ- lyase produced by Pseudomonas putida on several cell lines

The cytotoxicity of different concentrations of purified methionine γ- lyase from Pseudomonas putida on cancer cell lines (RD, AMN3 and AMGM) at 96 hr was studied. The bacterial enzyme with concentration 1000µg/ml was revealed highly cytotoxicity against cancer cell lines in comparison with other concentrations whereas slight cytotoxicity was observed on normal cell (REF).

Types of the Fiber Glass-Mat on Fatigue Characteristic of Composite Materials at Constant Fiber Volume Fraction: Experimental Determination

The aim of this work is to study the influence of the type of fiber glass –mat on fatigue behavior of composite material which is manufactured from polyester and E-glass (woven roving, chopped strand mat (CSM)) as a laminate with a constant fiber volume fraction (VF) of 33%. The results showed that the laminates reinforced with E-glass (woven roving) [0/90, ±45.0/90] and [0/90, CSM, 0/90] have lower fatigue strength than the laminates reinforced with E-glass [0/90]3,[CSM]3 and [CSM, 0/90, CSM]  although they had different tensile strength; the best laminate was [0/90]3 .
