In the present study, serum GOT(Glutamic Oxalacetic Transaminase) was purified, the purified enzyme showed the maximum activity at 37ºC and pH 7.5. During purification of serum GOT ion exchange chromatography lead to four separate forms (termed I, II, III and IV). GOT II with the highest specific activity was pure after chromatography on Sephacryl S300. . S.GOT levels were investigated in serum samples from patient with pulmonary tuberculosis .The S.GOT levels were determined at and 37ć .The study revealed that the serum GOT activity was higher in patient of tuberculous pulmonary than in control subjects.The mean serum GOT activity in the patients group was(90±8 IU/L) as compared to control group (27±0.65IU/L) showing a highly significant (p<0.001)difference .The study was concentrated to comprehensive measurement of the rate reaction kinetics(Km,Vmax,Hill coefficient),the first order forward rate constant k1 and half life t1/2 of the enzyme reaction in both normal and tuberculosis serum to evaluate any change In the number and structure of the active site then the rate of enzyme –substrate binding.

Nutrient agar medium with various concentrations of cefotaxime was used for isolation spontaneous mutants from wild type strain of P.aeruginosa PHA-1. Eighty-two mutants were successfully isolated with the viable count 52×107 , these mutants were confirmed as spontaneous not physiological adaption mutants by reculture on the same medium. Then, wild type PHA-1 and mutants were examined for production pyocyanin; a blue greenish pigment was clearly noticed on King A medium. Remarkably the mutant strain named S300-8 was distinguished in productivity in comparison with wild type strain PHA-1; the amount of pigment was 56.0667mg/l and 74.53mg/l respectively. The kinetic of atropine pertraction from seeds of Datura Metel Linn plant was studied. Diisopropyl ether, n-hexane and n-heptane were used as membranes for atropine recovery. The effect of speed of agitation and time in the range of 200-300 rpm and 0-3.5h, respectively were studied using the proposed membranes. The pertraction experiments were carried outs in a batch laboratory unit. The liquid-liquid pertraction was found to be very suitable for atropine recovery from its liquid extracts of Datura Metel seeds. A high purity (94-96%) can be obtained in the receiver phase.
