Isolation of fungi was performed from February to July, 2019. One hundred clinical specimens were collected from King Abdullah Hospital (KAH) Bisha, Saudi Arabia. Samples were collected from twenty patients of different ages (30 - 70 years old) ten males and ten females. The samples were collected from patients with the two types of diabetics. Specimens included blood, hair, nail, oral swabs and skin.  Specimens were inoculated on Sabourauds Dextrose agar containing chloramphenicol. Thirteen fungal species were isolated and identified. Mishrif Formation is the main reservoir in oil-fields (North Rumaila, South Rumaila, Majnoon, Zubair and West Qurna) which located at Basrah southern Iraq. A new spectrophotometric method for the determination of allopurinol drug was investigated. The proposed method was based on the reaction of the intended drug with catechol and Fe(II) to form a blue soluble complex which was measured at λmax 580 nm. A graph of absorbance versus concentration shown that Beer’s law was obeyed over the concentration range of 2–10 μg ml–1 with molar absorptivity of 9.4 x 103 l mol–1 cm–1 and Sandell sensitivity of 1.4 x 10–2 μg cm–2.
