This research deals with the color bias and its effect on maids in Mississippi in Kathryn Stockett''s (2003) The Help. The ill-treatment and negligence of Afro-American maids received from the white women who employed them in Mississippi that must have affected directly or indirectly on their personality and may eventually lead to suffering. They live in an atmosphere of struggle to free themselves from the complicated relationships between black and white. Afro-American maids pledged to liberate themselves from social oppression by protesting through writing a book which chronicles their stories in slave masters’ homes to make their presence felt as human being equal to their white masters.

This technique was broadly used in the late of nineteen century as a result of thedecay of plot, especially in novel writing. A number of aqueous samples were collected from river Tigris in Baghdad city, enriched ~1000 times using solid phase extraction (SPE), then extracted the trace concentrations of some polychlorinated biphenyls (PCB) using an aqueous two-phase system (ATPS) composed of 1Methylpyridinium chloride [MePy]Cl and KH2PO4 salt. In this study asparaginase was extracted from fruit part of Capiscum annum then Asparaginase activity was detected and optimised. Optimum conditions for the activity of crude asparaginase were studied. Results showed maximum activity of asparaginase was achieved 140 u/ml when the enzyme was incubated with 200 mM of asparagines at 35 °C for 30 minutes in the presence of 0.05 M of potassium phosphate buffer solution at pH 8.
