Determine the Biofilm Formed by Using ELISA Technology for Gram-Negative Bacteria Isolated from Wounds and Burns Infections, and the Study of the Production of the Biofilm Molecularly.

    This study was designed to investigate the capability of gram-negative bacteria that isolated form wound and burn infection to production of Biofilm which included (32) isolates, which have multi – drug resistant to antibiotics. The isolates included (10) Pseudomonas aeruginosa, (9) Klebsiella pneumoniae, (6) Escherichia coli, (5) Proteus mirabilis and (2) Enterobacter cloacae. The method used method links the crystal violet with biofilm and reading by ELISA which was adopted on the values of optical density of violets that linked to the mass of biofilm at the wavelength of (620) nm, the test results showed variation of biofilm composition for all bacterial species depending on the optical density value while the most production of biofilm was product form bacterial isolated from diabetes patients wound. Agarose gal electrophoreses results show that all isolate have a different plasmid profile according to the (size and position), it was also that temperature used as a curing factor causes a high frequency loss the plasmid from all isolates and according to this reason the yield was different after the curing that maybe refers to the role of both plasmid and chromosome in regulating this phenomena.

uropathogenic E.coli (UPEC)strains displayed a biofilm positive phenotype under
optimized condition using microtiter plate assay. 21 of E.coli strains classified as highly
positive biofilm producers (42%), and 29 (58%) as weakly positive biofilm producers.

Leuconostoc bacteria was isolated from local pickled cabbage (Brassica oleracea capitata) and identified as Leuconostoc mesenteroides by morphology,biochemical and physiological. This study included collection of 100 specimens from patients in AL-Kindy Teaching Hospital and teaching laboratories of Medical City Hospitals in Baghdad during the period from August to December 2012 ,these specimens differed in their sources which included 19 nasal swab, 16 wound swab,27 burn swab, 7 pus, 15 sputum, 10 corneal swab and 6 urine . Only 38 (38%) isolates was identified as Staphylococcus. In this study, 29 isolates (76.3%) were coagulase-positive (COPS), while only 9 isolates(23.6%) were coagulase negative (CONS), from total 38 isolates of Staphylococci.

Microalgae present much usefulness for antimicrobial research because of its enormous biodiversity and rapid growth rate. From this study results it is reaveled that Chlamydomonas reinhardtii were isolated from a pond of water in the province of Diwaniyah. The culture supernatants were obtained when extracted with methanol solvent. Antimicrobial activity of extracts was tested for pathogens, and the best inhibition zone obtained was against Candida albicans (32mm), S.aureus (15mm), and to E.coli (9mm). The aim of this research is to evaluate the effect of glucose and sodium chloride on biofilm formation by bacteria causing wound infection. For this purpose, 1% and 2% concentration of each of glucose and sodium chloride were used to test the biofilm formation potential of Staphylococcus aureus and Pseudomonas aeruginosa, which were the most common abundant bacteria that cause infection by biofilm.
