The Effect of Mercuric Exposure on Oxidative Stress and Enzymatic Antioxidant Defense System

Throughout the centuries, several incidents of mercury toxicity have been reported. Mercury is found in many industries such as battery, thermometer and barometer manufacturing, in the agricultural industry is used in fungicides and in medicine, mercury is used in dental amalgams. An important mechanism involved in cellular injury is induced by exposure to different forms of mercury involves in the induction of oxidative stress.      This study was conducted on non-smoker, male working in a chloroalkali plant for different periods, all workers were not suffering from chronic disease. Healthy non-smoker males that are not exposed, matched age were used as controls(C), workers aged (22-61) years, they were divided into three groups: G1: workers with exposed period less than 10 years, G1 < 10 years. G2: workers with exposed period (10-19) years . G3: included workers with exposed period more than 19 years, G3 > 19 years.     The result we had through examining the different parameters led us to add another group which included individuals with high mercury levels regardless the occupation period, in this group we found high significant changes in the defense system parameters that we measured.      This study showed an elevation in MDA levels in all workers group, specially those with high level mercury , which were 9.31, 12.78, 12.99, 14.73, and 18.11nmol/dl for C, G1, G2 , G3 ,and high level mercury workers respectively .      No alteration was found in SOD activity in erythrocyte (0.67, 0.73, 0.72, and 0.77 U/g.Hb) for C, G1, G2 and G3 respectively.       There were highly significant decrease in catalase activity (P < 0.001) in erythrocyte of all worker groups compared to normal control. The values were (1.5, 0.8, 0.88, 0.815 and 0.45 U/gm/Hb) for C, G1, G2, G3 and G4 respectively. While there were high elevation in glutathione S-transferase activity in worker groups compared to control (P < 0.001) and values were (1.85, 2.589, 2.441, 2.776 and 3.2 U/gm.Hb) for C, G1, G2 and G3 respectively.

The Effects of Melatonin On The Oxidative Stress , Protein Glycation , Microalbuminuria and Lipid Profile In Type II Diabetes Mellitus

Previous studies indicated that supplementation with antioxidants has a protective effects against oxidative stress–induced damage in type 2 diabetes. In this study we evaluated the antioxidant effects of melatonin on the oxidative stress parameters and microalbuminuria in type 2 DM patients. 30 patients with type 2 DM were treated with 3mg/day melatonin for 90 days. Erythrocytes and plasma MDA and glutathione, fasting plasma glucose, %HbAIC, microalbuminuria, total plasma protein and lipid profile were measured each 30 days and compared with those obtained from 20 healthy controls.

A decrease in MDA levels associated with the elevation in GSH levels were observed, compared with the pre–treatment levels.

Oxidative Stress in Multiple Sclerosis Disease (Review Article)

Background: Multiple sclerosis (MS) is an inflammatory disease of the central nervous system, in which the myelin sheaths got injured. The prevalence of MS is on grow, as well as, it affects the young ages. Females are most common to have MS compared to males. Oxidative stress is the situation of imbalance between oxidants (free radicals and reactive oxygen species (ROS)) and antioxidants in a living system, in which either the oxidants are elevated or antioxidants are reduced, or sometimes both. ROS and oxidative stress have been implicated in the progression of many degenerative diseases, which is important in cracking the unrevealed mysteries of MS. In this review article, some of the proposed mechanisms that link oxidative stres

Purification of Fungul Glucoamylase and Study the Effect of Concentration And Thermal Processing on Enzymatic Activity of Starch Hydrolysis

Glucoamylase from black Aspergillus niger isolate was purified by ammonium sulfate precipitation and sephadex G 200 filtration. A trial for the purification of glucoamylase resulted in an enzyme with a specific activity of 6472 unit/mg protein with 10 times fold. The main goal of the present work was to test this enzyme in hydrolyzing the raw starch. Two substrates were used: corn starch and potato starch 2% (w/v). The effect of enzyme dosage and thermal processing of substrate on kinetics and efficiency of hydrolysis were studied. The results suggested that the glucoamylase activity is increased as the increase of enzyme concentration and the enzyme was sufficiently effective in hydrolyzing tested raw starch, and thermal modification of

UV-Exposure effect on the mechanical properties of PEO/PVA blends

Poly ethylene oxide PEO / Poly vinyl alcohol PVA  blends were prepared by cast method at different ratios of (25%PEO/75%PVA, 30%PEO/70%PVA, 35%PEO/65%PVA, 40%PEO/60%PVA, and 50%PEO/505PVA). Samples miscibility ,and thermal stability were studied  by using differential scanning calorimetry(DSC),and thermo gravimetric analysis (TGA) analysis. The results proved that there was one glass transition temperature (Tg=160°C) at 25%PEO\PVA ratio
)that was attributed to its miscibility. This miscibility associated with (Hydrogen  bonds) between (Hydroxyl group) in PVA, and PEO, whereas there were two glass transition temperature for the blends ratio more than 30%PEO,that was due its immisc

Serum Oxidative Stress and Creatine Kinase Activity Related to Hoffman's Syndrome

Several symptoms of hypothyroidism characterized by low

FT3 and FT4 were classified into hypothyroidism and Hoffman's syndrome .It was accepted that free radical generation and lipid peroxidation are associated with  many diseases. Creatine kinase in serum is affected by thyroid hormones. This study was undertaken to investigate the relationship between  malondialdehyde  as a general lipid peroxidation marker and CK activity in patients with primary hypothyroidism and Hoffman's syndrome which indicatees hypothyroidism with undetectable FT3 and FT4 in serum with elevated values of  TSH.

The study revealed that MDA is within the normal range, while CK activities in sera of Hoffman's syndrome were increas

Biomarkers of Oxidative Stress in Diabetic Microvascular Complications Review Article

Reactive oxygen species (ROS) are produced as a result of biochemical processes that are not in balance with the body's antioxidant defense mechanism. This metabolic dysfunction is referred to the oxidative stress (OS).  Metabolic dysfunction-associated diseases are affected by changes in the redox balance. It is now widely recognized that oxidative stress significantly affects diabetes mellitus (DM), particularly type 2 diabetes. The biochemical changes associated with  DM could  disturb the oxidative milieu, leading to several microvascular complications in diabetic patients. Thus, DM is a perfect disease to explore the harmful consequences of oxidative stress and how to treat it. Oxidative stress triggered by hyperglycemia is
