Detection of Transcription Factor C-MYC in Oral Dysplasia and Squamous Cell Carcinoma by in Situ Hybridization

Background: Oral squamous cell carcinoma continues to be a major health problem in Iraq as well as in other countries. Many attempts were made to study molecular markers in oral squamous cell carcinoma and to link them to tumor grade, stage and prognosis, besides studying their role in carcinogenesis. The present study has been designed to detect mRNA of c-myc in oral squamous cell carcinoma compared to oral dysplasia and to link the marker to grade and degree of the two pathologies.
Materials and methods: Forty two cases, including 30 cases of oral squamous cell carcinoma and 12 cases of oral dysplasia were included in this study. Sections on positively charged slides were made from their paraffin blocks and were used for the detection of c-myc mRNA using in-situ hybridization technique.
Results: C-myc mRNA was detected in 9(75%) cases of oral dysplasis and in 24 (80%) cases of oral squamous cell carcinoma. A significant correlation was found between c-myc mRNA score and intensity from one side and the tumor grade from the other side and degree of dysplasia.
Conclusion: The results of this study confirm the role of c-myc in oral dysplasia and oral squamous cell carcinoma and the possible transfer from the former to the latter.

Expression of TIMP-2 in HPV-16 infected oral squamous cell carcinoma in patients in Iraq

Aim: To determine the expression of tissue inhibitors of metalloproteinases (TIMP-2) in oral squamous cell carcinoma (OSCC) and the difference in its expression level between positive and negative HPV-16 (human papilloma virus- 16) OSCC patients. Methods: This study was conducted on 33 biopsies obtained from patients with OSCC and 10 normal oral mucosa as controls. In situ hybridization (ISH) was used to investigate the presence of HPV-16, while immunohistochemistry (IHC) was used to estimate the expression level of TIMP-2. Results: The TIMP-2 was expressed in 27 (81.8%) of OSCC sections with no significant difference between its expression level in HPV-16 positive and HPV-16 negative OSCC cases (p=0.058). TIMP-2 was found to be hig

Immunohistochemical expression of Fas/Fasligand and c-Myc in oral lichen planus

Background: Oral Lichen Planus is a chronic inflammatory mucosal disease, presenting in various clinical forms .Both antigen-specific and non-specific mechanisms involved in the pathogenesis of OLP. Apoptosis or programmed-cell death is a physiological process essential for the normal development and maintenance of homeostasis in many organisms. Fas is a cell-surface glycoprotein, 40-kDa, that belongs to the nerve growth factor / tumor necrosis factor (TNF) receptor family. Fas is expressed in several tissues including blood, where its expression is upregulated on activated T and B lymphocytes and natural killer cells. Fas ligand is a type II transmembrane protein that belongs to the tumor necrosis factor family. The proto-oncogene c-Myc is

P16 Protein and Human Papillomavirus (HPV16, 18) Expressions in Oral Lichen Planus and Squamous Cell Carcinoma

Background: Oral carcinogenesis is a molecular and histological multistage process featuring genetic and phenotypic markers for each stage, which involves enhanced function of several oncogenes and/or the deactivation of tumor suppressor genes, resulting in the loss of cell cycle checkpoints. The progression towards malignancy includes sequential histopathological alterations ranging from hyperplasia through dysplasia to carcinoma in situ and invasive carcinoma. The p16 gene produces p16 protein, which in turn inhibits phosphorylation of retinoblastoma, p16 play a significant role in early carcinogenesis. Human papillomavirus is a well established heterogeneous virus and plays an important role in oral cancers. The aims of the study were to

Immunohistochemical expression of Cyclin D1 and NF-KB p65 in oral lichen planus and oral squamous cell carcinoma (Comparative study)

Background: Oral Lichen Planus (OLP) is a chronic inflammatory mucosal disease, presenting in various clinical forms WHO had regarded OLP as a precancerous conditions in 1978 because of its potential with cancer. Both antigen-specific and nonspecific mechanisms involved in the pathogenesis of OLP. Oral Squamous Cell Carcinoma (OSCC) is the most common malignant neoplasm of the oral cavity representing more than 94% of oral cancer. It occurs in different sites and has many etiological factors. Cyclin Dl is a proto-oncogene which consider as the key protein in the regulation of cell proliferation and its overexpression led to the occurrence and progression of malignant tumors.NF-KB p65 is a member ofNF-kB family of transcription factors that

Detection of human cytomegalovirus genome in malignant gliomas by in situ hybridization technique

Background: Human Cytomegalovirus ( HCMV ), lies dormant in the glial cells , and can be reactivated under conditions of inflammation and immunosuppression. In vitro, the virus can
transform cells and dysregulate many cellular pathways involved in oncogenesis. This study was conducted to detect HCMV matrix-protein mRNA using In Situ Hybridization technique in glial brain tumor tissues compared to normal brain tissues and the presence of cytomegalic inclusion bodies in brain tumor tissues.
Patients and Method: Thirty eight of glial tumor specimens were obtained in paraffin blocks compared to eight normal brain autopsy specimens which were age and sex matched with the
study group as a control group. ISH wa

A Salivary Calcium Binding Protein in Patients with Oral Squamous Cell Carcinoma in Relation to Smoking

Background: The present study aimed to assess the distribution, prevalence, severity of malocclusion in Baghdad governorate in relation to gender and residency Materials and Methods: A multi-stage stratified sampling technique was used in this investigation to make the sample a representative of target population. The sample consisted of 2700 (1349 males and 1351 females) intermediate school students aged 13 years representing 3% of the total target population. A questionnaire was used to determine the perception of occlusion and orthodontic treatment demand of the students and the assessment procedures for occlusal features by direct intraoral measurement using veriner and an instrument to measure the rotated and displaced teeth. Molecular and Phylogenetic Analysis of Human Papillomavirus Using L1 Gene in Oral Squamous Cell Carcinoma Patients in Baghdad, Iraq

Oral squamous cell carcinoma (OSCC) is the most common malignant neoplasm of the oral mucosa. Human papillomavirus (HPV) virus cause a broad scope of diseases from benign to invasive tumors, types 16 and 18 classified as carcinogenic to humans. This study aimed to provide the first molecular characterization of HPV types in Iraq. Thirty-five unstimulated whole saliva samples were collected from histopathologically confirmed patients with oral cancer were enrolled in this study. Genomic DNA was extracted from exfoliating cells to amplify HPV-DNA using HPV-L1 gene sequence primers by polymerase chain reaction method (PCR), the viral genotyping was performed using direct sequencing method. HPV genotypes identified were deposited in Gen

β- catenin Expression and Its Relation to Bryne’s Invasive Grading System in Oral Squamous Cell Carcinoma

Background: Invasion in oral cancer involves alterations in cell-cell and cell-matrix interactions that accompanied by loss of cell adhesion. Catenins stabilize cellular adherence junctions by binding to E-cadherin, which further mediates cell-cell adhesion and regulates proliferation and differentiation of epithelial cells. The Wnt/β-catenin pathway is one of the major signaling pathways in cell proliferation, oncogenesis, and epithelial-mesenchymal transition. Aims of the study: to detect immunohistochemical distribution pattern and different subcellular localization of β-catenin in oral squamous cell carcinoma and relate such expression to Bryne’s invasive grading system. Materials and Methods: This study included 30 paraffi
