P. aeruginosa is a famous bacterium that causes several diseases and has a high ability to be a multidrug resistant organism that is linked with the formation of biofilm. This study aimed to investigate tssC1 gene role in the resistance of different antibiotics in the presence of biofilm. We constructed biofilm for the isolates under the study and showed the effect of different antibiotics on biofilm formation and maturation. The presence of the gene was detected through achieving PCR reaction. Finally, tssC1 gene variation was determined through sequencing and aligning the sequencing products. The results showed that most of the isolates (80%) formed biofilm that played a role in the resistance of different antibiotics which could be due to the presence of tssC1 gene. However, the genic variation of tssC1 gene showed that no variation was detected. Therefore, we think this gene has no a role in the resistance of antibiotics and that the resistance may have been raised by other mechanisms found in P. aeruginosa isolates. This led us to conclude that the tssC1 gene does not contribute to the resistance of antibiotics through biofilm.
Pseudomonas aeruginosa was isolated from various clinical samples included urine, sputum, stool, ear, wound & burn swabs. Detection of the ability of local isolates to produce staphylolysin enzyme was studied, on Tryptic soya agar + 0.2% (wt./vol.) of heat killed Staphylococcus. aureus at temperature 100oC. medium and the diameters of lysis zone ranged from 5-22mm, then the isolate P16 was chosen to extract staphylolysin A (LasA) and its specific activity reaches 8.59 unit /mg protein, whi1e the isolate P5 was chosen to extract staphylolysin D (LasD) where it's specific activity reaches 0.66 unit /mg protein since the two isolates were the most production of enzyme. Staphylolysin enzyme was extracted by cooling centrifugation and par

Objective: The present study aimed to evaluate the antibacterial activity of L. Sativum seed extract against multi drug resistant (MDR) and sensitive Pseudomonas aeruginosa clinical isolates.
Subjects and Methods: An ethanolic and aqueous stock extracts were prepared from L. sativum seed plant then serial dilutions were prepared and the obtained concentrations (50, 25, 12.5 and 6.2 mg/ml) were tested against 30 multidrug-resistan
... Show MoreOne hundred isolates of Pseudomonas aeruginosa were obtained from patients admitted to Baghdad hospitals, Iraq during the period between May 2018 until July 2018. These isolates were distributed as 15 isolates from blood, 25 isolates from urinary tract infections, 10 isolates from sputum, 12 isolates from wounds, 15 isolates from ear infections, 15 isolates from bronchial wash of patients suffering from respiratory tract infections in addition to 8 isolates from cystic fibrosis patients. The isolates were initially identified by culturing on MacConkey agar, blood agar and P. aeruginosa agar then diagnosed by performing some morphological and biochemical tests. The second diagnosis was done by API 20E system followed by Vitek 2 compact syste

aeruginosa isolated from eye infection. One hundred samples (corneal scrapings)
were obtained. Approximately, 77% of the cases were due to contact lens wear
followed by 15 % trauma and 8% with unknown history. The isolates identified as
P. aeruginosa were 30% (23/77 CL) and 25% (2/8 Unknown). On the other hand,
the Kirby-Bauer antibiotic sensitivity assay showed that 100% of the isolates were
sensitive to Neomycin, Gentamicin and Amikacin. While 91.6% were sensitive to
Carbenicillin and Ceftriaxone; 66.6% were sensitive to Cefotaxime and 0% were
sensitive to Tertacycline. Only two isolates were found to be multidrug resistant.
Screenin
Pseudomonas aeruginosa gram-negative, bacilli and facultative aerobic, P. aeruginosa cause cystic fibrosis patients, wounds, burns, and immunodeficienct patients, that have many virulence factors such as pyocyanin , cytotoxic ,biofilm formation and motility, Eighty-eight isolates belonging to P. aeruginosa were collected including the 66 clinical isolates obtained from different hospitals in Baghdad and were from different sources and 22 environmental isolates from previous studies of soil near oil fields. Microscopical and cultural characteristics were studied and diagnosed using biochemical tests, VITEC device, their ability to adhere to non-living (Polystyrene), living cell line (A549) and cytotoxicity of bacterial filtrate

Bacterial isolates were identified using conventional biochemical tests and then identification was confirmed by using Vitek-2 compact system. Pseudomonas aeruginosa recovery was 60 isolates in this study. These isolates were analyzed for antibiotic susceptibility by the disk diffusion test (DDT) according to Kirby Bauer's method using seven clinically important antipseudomonal agents: carbapenems (Imipenem and Meropenem), pencillins (Piperacillin), cephalosporins (Ceftazidim), monobactam (Aztreonam), quinolones (Ciprofloxacin) and aminoglycosides (Gentamicin). Show MoreThe study included the collection of samples of raw cow milk to isolate Leuconostoc bacteria, samples were sub cultured on De-Man Rogosa Sharpe-Vancomycin medium, the pure colonies were selected and subjected to the cultural and microscopically tests, according to that 25 cocci bacterial isolates were obtained, then isolates were subjected to biochemical tests. Result of tests showed that 12 isolates belong to the genus Leuconostoc out of 25 cocci bacterial isolates, Vitek2 system was used as a supplementary step. Results of final identification showed that 3 sub species were obtained included Leuconostoc mesenteroides ssp. cremoris 9 out of 12 isolates, while it was 2 isolates of Leuconostoc mesenteroides ssp. mesenteroides and one isol

In this study, detection of uricase production from Pseudomonas aeruginosa
isolates was done by applying colorimetric method, Uricase was purified from the
most potent isolate by precipitation using ammonium sulphate (80% saturation) then
purification was achieved using DEAE –Cellulose ion exchange and Sepharose 6B
gel filtration chromatography column, 16.4% of total enzyme was recovered with
specific activity 2337.5U/mg and 22.21folds of purification. Characterization of
uricase involved detection of optimal conditions for uricase activity, the maximal
activity was obtained at temperature 45ºC,while uricase appeared to be stable at
40ºC. Uricase showed optimal activity at pH 9 while pH stability was in the
