Resistance to aminoglycosids is a great problem to therapeutics. Aminoglycoside acetyltransferase producing Pseudomonas aeruginosa have been reported to be important cause of nosocomial infections. The purpose of this study was to determine the occurrence of aminoglycoside acetyltransferase. A total of 200 clinical and environmental samples were collected over period of five months. The P. aeruginosa isolates were confirm their identification, antibiotic susceptibility profile according to vitek2 compact system. The isolates were subjected to polymerase chain reaction (PCR) assays with specific primers for aac (6')-I, aac (6')-Ib, aac (3')-I . Only 32 (16.%) P. aeruginosa isolates were recovered from the samples. in present investigation. Gentamicin seemed to offer more resistance (31.3%) than tobramycin (28.1%), which itself is slightly more resistance than amikacin (25%).In PCR experiments using specific primers for genes. aac(6')-I , and aac(6')-Ib were present in 12 (37.5%) and 15 (46.9%) of the isolates, respectively. While the aac (3')-I were negative among all isolates.
Fifteen local isolates of Pseudomonas were obtained from several sources such as soil, water and some high-fat foods (Meat, olives, coconuts, etc.). The ability of isolates to produce lipase was measured by the size of clear zone on Tween 20 solid medium and by measuring the enzymatic activity and specific activity. Isolate M3 (as named in this study) was found to be the most efficient for the production of the lipase with enzymatic activity reached 56.6 U/ml and specific activity of 305.94 U/mg. This isolate was identified through genetic analysis of the 16S rRNA gene. and it was shown that the isolate M3 belongs to Pseudomonas aeruginosa with 99% similarity. The DNA of isolate M3 was extracted and lipase gene was amplified through PCR tec
... Show MoreOwing to high antibacterial resistance of Pseudomonas aeruginosa, it could be considered as the main reason behind the nosocomial infections. P. aeruginosa has a well-known biofilm forming ability. The expression of polysaccharide encoding locus (pelA gene) by P. aeruginosa is essential for this ability. The purpose of the current research was to determine the biofilm formation in P. aeruginosa isolated from clinical samples and to evaluate the role of the selected PelA gene in biofilm formation using PCR method in Iraqi patients. Results revealed that 24 (96%) isolates were found to have the ability to form biofilm that was remarkably related to gentamicin resistance. Moreover, the pelA gene was found in all biofilm-producers. In conclu

Show MorePseudomonas aeruginosa is a common and major opportunistic human pathogen, its causes many and dangersinfectious diseases due to death in some timesex: cystic fibrosis , wounds inflammation , burns inflammation , urinary tract infection , other many infections otitis external , Endocarditis , nosocomial infection and also causes other blood infections (Bacteremia). thereforebecomes founding fast and exact identification of P. aeruginosafrom samples culture very important. However, identification of this species may be problematic due to the marked phenotypic variabilitydemonstrated by samples isolates and the presence of other closely related species. To facilitate species identification, we used 16S ribosomal DNA(rRNA) sequence data
... Show MoreAccording to the prevalence of multidrug resistance bacteria, especially Pseudomonas aeruginosa, in which the essential mechanism of drug resistance is the ability to possess an efflux pump by which extrusion of antimicrobial agents usually occurs, this study aims to detect the presence of mexB multidrug efflux gene in some local isolates of this bacteria that show resistance towards three antibiotics, out of five. Sensitivity test to antibiotics was performed on all isolates by using meropenem (10µg/disc), imipenem (10µg/disc), amikacin (30 μg/disc), ciprofloxacin (5µg/disc) and ceftazidime (30 µg/disc). Conventional PCR results showed the presence of mexB gene (244bp) in four isolates out of t
... Show MoreThe current study includes 144 samples were 106 bacterial samples belonging to the clinical sources, 38 bacterial samples belonging to the environmental sources to investigate the presence of bacteria P. aeruginosa. The results of diagnosis clarified that there are 45 bacterial isolates belonging to the bacterium P. aeruginosa The examination of the sensitivity of all bacterial isolates was done for elected 45 isolation towards the 11 antibiotic by spread method on the dishes. The results showed that the resistance ratio toward Cefixim, Cefotaxim, Tetracycline, Amoxicillin, Cloxacillin, Methicillin, Erythromycin and Naldixic acid was 77.7, 73.3, 84.4, 82.2, 80, 77.7, 77.7 and 73.3 respectively, While most isolates were sensitive to all o
... Show MoreIn this study, 158 clinical samples were collected from hospitalized burn patients during the period from December 2012 to June 2013 in Karbala province\ Iraq. Bacterial isolates were identified using conventional biochemical tests and then identification was confirmed by using Vitek-2 compact system. Pseudomonas aeruginosa recovery was 60 isolates in this study. These isolates were analyzed for antibiotic susceptibility by the disk diffusion test (DDT) according to Kirby Bauer's method using seven clinically important antipseudomonal agents: carbapenems (Imipenem and Meropenem), pencillins (Piperacillin), cephalosporins (Ceftazidim), monobactam (Aztreonam), quinolones (Ciprofloxacin) and aminoglycosides (Gentamicin). Show MoreSeparation of uricase from Pseudomonas aeruginosa was done using (70%) satu-ration ammonium sulphate, and purification of this enzyme was done by ion ex-change chromatography on DEAE- cellulose column and eluted with linear NaCl (0-1M). Partial purified uricase gave an activity of (4.9 u/ml), protein concentration of (0.56 mg/ml), specific activity of (8.75 unit/mg) with purification folds (8.4) and a yield of (48%). The maximum purified uricase activity was detected at 35ºc and pH 8.5 with (0.12 mM.uric acid). The results shown that red cabbage extract (RCE) contain flavonoides which contain phenolic compounds and anthocyanines which glycoslated with mono or dimolecules of saccharides, while test for alkaloids, ster-oids, saponins and
... Show MoreOut of 120 isolates from different clinical cases, only 75 were found and confirmed that they belong to the Pseudomonas aeruginosa bacteria. The result revealed that the LasB virulent gene was present in 63 isolates with 63% percentage. The gel electrophoresis showed that the molecular weight of LasB gene was 300 bp. DNA sequences of LasB gene was done, and the results showed the presence of some gene mutations like substitution, addition and deletion with 97% identity with the Refseq gene. From the other side, the results of identities of translated nucleotides sequence with the original sequence of amino acids revealed that there are no effects of gene mutations on translation of the product protein.
