The present study was aimed to screen the ability of local isolates of Bacillus spp. (56 isolates) for nattokinase production using solid state fermentation, then optimize the nutritional conditions for enzyme production. The isolates were subjected to the primary and secondary screening process to select the Bacillus isolate which give the highest production of enzyme. It was found that Bacillus sp. B24 had the highest productivity of the enzyme (25.58U/mg protein). The optimum conditions for nattokinase production were performed by the solid state fermentation and found that the wheat bran was the best medium at initial moisture ratio 1.0:1.0 (w/v) using distilled water as moisturizing solution with initial pH of 7.0 after inoculation with 5×107 cell/ml for 72 hrs, the enzyme specific activity was 38.65U/mg protein.
Backround: The Solid state fermentation has several advantage including absence of free water , reduced volume of production media utilized for high products and the relatively low costs of production.
Methods: Thirty local isolates of soil obtained from Genetic Engneering and Biotechnology Institute. Nutrient agar was used to growth strains examination to antibacterial agent and Wheat bran and fish meal were used in combination (0-100%of each )and divided in 10 gm lost /flask . Each flask is inoculated with different numbers of Streptomyces spores and incubated for 5 days at 28°C, then the supernet was extracted and were assayed as antibacterial
Results: The ability of 30 local isolates of Streptomyc
Lovastatin is one of the most important compounds that is produced from some filamentous fungi, being employed in the reduction of hypocholesterolemia. The results of screening, after the collection of seventy-three local fungal isolates from different areas, demonstrated that the local isolate Aspergillus terreus A50 was the best isolate for lovastatin production, with a concentration of 12.66 µg/ml, through the submerged fermentation. Lovastatin produced from A. terreus A50 showed antimicrobial activities against a Candida albicans isolate. Solid state fermentation (SSF) was the best system to produce the highest yield of lovastatin by A. terreus A50 as compared to the submerged fermentation (SmF)

Show MoreThe study includ selection of six species of the fungi related to genus Pleurotus were evaluated for their ability to produce of Pleurotin, one of them, Pleurotus ostreatus (P.11) was isolated and identified in the present study. Pleurotin was detected by Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC). The maximum absorption of Pleurotin was 1.6 nm at 250 nm. Pleurotin was purified with two methods using chloroform and ethyl acetate, the results showed the ethyl acetate was more efficient in pleurotin production resulting in 14.6 μg/ml compared to 9.8 μg/ml with chloroform. The local isolate, P. osteratus (P.11) showed significant high Pleurotin production (14.6 μg/ml) when was grown on the modified

plant roots. The ability of inulinase production by these isolates was screened.
Bacillus Be9, which isolated from soil of lettuce root, was the highest inulinase
producer; it was identified as Bacillus cereus. Optimal culture medium and
condition for inulinase production were determinatd; the highest inulinase
production was obtained when the bacteria was cultured in inulin medium which
contained 0.5% inulin, 0.4% peptone as carbon and nitrogen source at pH 7.0
inoculated with 1ml of bacterial suspension and incubated at 40˚C for 48hrs.
The pollution producing from textile industries effluents is growing since the years, due to at discharged lots of it in water without treatment. The resulting effluent is colourful, highly toxic, and poses a significant environmental hazard. This problem can be solved by using enzymic biological treatment, where the Congo red dye was used with concentrations (100,200,300,500) mg /L, pH values (3,4,5,6,7,8), and variable temperatures (25,35,45)°C, the best removal of Congo red (CR) dye under optimum conditions for degradation was at concentration of 100 mg/L, at (pH 6, 25 °C) with efficiency of 99.85 % using the peroxidase enzyme extracted from red radish plant, while the removal percentage decreased when increase dye concentration

We studied the effect of certain environmental conditions for removing heavy metal elements from contaminated aqueous solutions (Cd, Cu, Pb, Fe, Zn, Ni, Cr) using the bacterium Bacillus subtilis to appoint the optimal conditions for removal ,The best optimum temperature range for two isolate was 30-35○C while the hydrogen number for the maximum mineral removal range was 6-7. The best primary mineral removal was 100 mg/L, while the maximum removal for all minerals was obtained after 6 hrs of Cu element time and the maximum removal efficiency was obtained after 24 hrs of Cu element. The results have proved that the best aeration for maximum removal was obtained at rotation speed of 150 rpm/minute. Inoculums of 5ml/100ml which contained 1

These conditions are implemented to test the effect of the variables of the process of hydrolysis on the nature and quality of the product.
The production of soya peptone was studied for their importance in the process of preparing and producing the culture media used in medical and microbiological laboratories.
The process of production of soya peptone includes four main
