The Production of Biofilm from Methicillin Resistant Staphylococcus Aureus Isolated from Post-Surgical Operation Inflammation

Methicillin resistant Staphylococcus aureus (MRSA) is the most common pathogenic bacteria in the hospitals and communities, the ability to form biofilm is considered the main cause of Staphylococcus pathogenicity since it provides resistance to both antibiotics and host immune response, so this study was aimed to evaluate the biofilms formation and its association with antibiotic resistance in clinical isolates of MRSA, in order to achieve this aim, 237 samples were collected from different patients with wounds infections after surgeries and samples from operations galleries from varies hospitals in Baghdad ,68 isolates out of 237 were subjected to Staphylococcus aureus according to conventional methods Additionally, the S. aureus isolates were re-identified by molecular method. The results for Susceptibility of 68 S.aureus isolates against 22 antimicrobial agents revealed all isolates were resistant (100%) to Cefotaxime, and showed resistance to Ceftriaxone, Oxacillin, Cefipime, Cefoxitin, ampicillin, Penicillin, Nalidixicacid, erythromycin in percentage (99, 97,97,87, 87, 79,  75,   65  )%  respectively while other isolates showed variety in their resistant to other antimicrobial agents. while all 68 isolates were sensitive to Nitrofurantoin which is considered the most effective agent against the isolates. All 68 isolates were identified as MRSA on HiCromeMeReSa Agar Base medium, but 66 among  68 isolates were methicillin resistant in antibiotic sensitivity test, while 67 isolates had a positive reaction when detecting mecA gene using Polymerase Chain Reaction (PCR) technique.  The finding of biofilm detection by microtiter plate method (MTPs) showed that 56 of 68 isolates produced biofilm in different degrees, while 66 (97%) among 68 isolates possessed the icaAD gene using PCR.

Bacteriocin Typing of Staphylococcus aureus Isolated from Different Sources in Ibb City, Yemen

Abstract A total of 207 specimens were collected from different sources including patients, health care staff and hospital environment in Ibb city, Yemen. The study used the bacteriocin produced from active producer strains in typing of Staphylococcus aureus. Depending on the morphological, cultural and biochemical characteristics, 54 (26.09%) isolates of Staphylococcus aureus were identified. An antibiotic sensitivity test was done for the bacterial isolates, and the results showed that there were multiple resistant antibiotics. The Staphylococcin production of these isolates has been detected by using wells assay. Fifty one isolates were Staphylococcin producer. Four isolates (staph19, staph25, staph28 and staph43) were chosen as go

The Correlation Between the Persistence of Methicillin Resistant Staphylococcus Aureus Isolates to Mupirocine and Toxin-Antitoxin Type II Genes

Under high concentrations of antibiotics, a fraction of the bacterial population exhibits a phenomenon known as persistence. Toxin- system (TA system) has been reported to be involved in the formation of E. coli, Mycobacterium, and S. aureus persisters.  In this study, the ability of thirty Iraqi isolates of MRSA to form in vitro persister cells after exposure to three different antibiotics (Ceftriaxone 30 µg, Mecillinam 10 µg, and Mupirocin 20 µg) was examined by TD test. Additionally, efflux pump inhibitor [Fluphenazine 0.25 mg/ml] was combined with the antibiotic that triggered persister formation. The distribution of mazEF and yefM-yoeB (Type II TA system) in the tested isolates was detected by PCR. 91% of Mupirocin su

EFFECT OF CHALCONE ON THE FORMATION OF BIOFILMS AND EXPRESSION OF VIRULENCE GENES IN METHICILLIN-RESISTANT STAPHYLOCOCCUS AUREUS

MRSA is one of the major pathogens in hospitals and the community, which have the ability to produce biofilm as a virulence factor, the impact of chalcone on biofilm formation, the synergism effect of chalcone and antibiotic in both in vitro and in vivo experiments, the gene expression of virulence genes (srtA, fnbA, fnbB) before and after treatment of it on MRSA biofilm cells in vitro, all these were the prime aims of this study. Chalcone at MBIC (20 μg/ml), significantly reduced the biofilm formation to 21.45% and at sub MBIC (15 μg/ml) to 36.58 %. While, Chalcone at MIC(5 μg/ml) reduced MRSA planktonic cells to 49.61%. Susceptibility of MRSA isolates against eight antibiotics showed that all isolates were sensitive to vancomycin and n

Comparison between Cefoxitin disk diffusion, Crome agar and EPI-M Screening Kit for Detection of Methicillin- Resistant Staphylococcus aureus

Specimens have been collected from one hundred and seventeen patients residing in local hospitals, 33 with burns and 84 wound injuries,. Three different methods ,Cefoxitin disk diffusion, EPI-M Screening Kit and Crome agar (MeReSa agar)with selective supplement were used to detect methicillin-resistant Staphylococcus aureus (MRSA) . A comparison was made between these 3 methods according to the results. It was found that the results of the Cefoxitin disk diffusion test were compatible with the results of culturing on Crome agar, while those obtained from the EPI-M Screening kit were not accurate and some of them gave false negative results.
