Bacteriological and Molecular Study of Fluoroquinolones Resistance in Pseudomonas aeruginosa Isolated From Different Clinical Sources

The present study was conducted to investigate the resistance of fluoroquinolones (FQs) and the effects of mutations in the resistance gene in clinical isolates of P. aeruginosa isolated from different sources in Al-Hussein Hospital, Al-Samawah city, Iraq. The basic mechanism of the resistant of fluoroquinolones in P. aeruginosa is via mutations occurring in the basic bacterial gyrA gene encoding-subunit A of DNA gyrase . Forty clinical isolates from various sourced  (burn 7 (17.5 %), wound 7 (17.5 %), ear 2 (5 %), operation room 12 (30 %), urine 3 (7.5 %), and industrial dialysis center 9 (22.5 %)) were isolated based on bacteriological methods confirmed by 16s rRNA gene using PCR technique. A sensitivity test was conducted to all isolates by Kirby-Pour method using 7 antibiotics of fluoroquinolones. Amongst the 40 clinical isolates, 10 were resistant and 3 were sensitive to all tested antibiotics, while 27 were intermediate, resistant and sensitive to two or more of tested antibiotics, with the resistance being confirmed by the minimum inhibitor concentration (MIC) test. The ten resistant isolates were used to examine the mutations in gyrA gene. A direct sequence method was used and revealed eight mutations in gyrA gene at different positions. In addition, we found that fluoroquinolone activity in the sensitive isolates, after sequencing for these isolates, is a bacteriostatic activity. The results of this study showed the gyrA mutations resulting from the excessive use of antibiotics are one of the mechanisms may be that leading to fluoroquinolone resistance.

Isolation and Identification of Novel Streptococcus pluranimalium Isolated from Children with Upper Respiratory Infections in Mosul Hospitals, Iraq

Streptococcus pluranimalium was first isolated in 1999. Recently, several case reports have been published that have revealed that S. pluranimalium can infect humans as well. The pathogenicity and pathophysiology of this pathogen is poorly studied and its characteristics are not well known. In this study, S. pluranimalium was first isolated and then identified from infants and children who suffered from upper respiratory infections. 90 samples were collected from nasopharyngeal cavity. Among them, 83 Streptococcus spp. isolates were identified. 3 out of which were biochemically and molecularly identified as S. pluranimalium. 16S rRNA sequencing based identification revealed that all iso

A Developed Colorimetric Assay Using Unmodified Gold Nanoparticles for the Identification of Acinetobacter baumannii Isolates from Different Clinical Samples

Acinetobacter baumannii (A. baumannii ) is considered a critical healthcare problem for patients in intensive care units due to its high ability to be multidrug-resistant to most commercially available antibiotics. The aim of this study is to develop a colorimetric assay to quantitatively detect the target DNA of A. baumannii based on unmodified gold nanoparticles (AuNPs) from different clinical samples (burns, surgical wounds, sputum, blood and urine). A total of thirty-six A. baumannii clinical isolates were collected from five Iraqi hospitals in Erbil and Mosul provinces within the period from September 2020 to January 2021. Bacterial isolation and biochemical identification of isolates

Study of Molecular Interaction for Antibiotic Drug with Sugar Solutions at Different Temperature

The interactions of drug amoxicillin with maltose or galactose solutions with a variation of temperature have been discussed by taking in the volumetric and viscometric procedures. Physical properties [densities (ρ) and viscosities (η)] of amoxicillin (AMOX) aqueous solutions and aqueous solutions of two type saccharides (maltose and galactose 0.05m) have been measured at T = (298.15, 303.15 and 308.15) K under atmospheric pressure. The apparent molar volume (ϕv cm3mole-1) has been evaluated from density data and fitted to a Redlich-Mayer equation. The empirical parameters of the Mayer-Redlich equation and apparent molar volume at infinite dilution Ø°v were explicated in terms of interactions from type solute-solvent and solute

Developing multiplex PCR for the rapid and simultaneous detection of Staphylococcus aureus, Pseudomonas aeruginosa and Klebsiella pneumoniae associated with sheep respiratory tract infections

Respiratory tract infections in sheep are among the important health problems that affect all sheep ages around the world. Nine bacterial isolates obtained from sheep with respiratory tract infections were selected to be used in the current study. The isolates included 3 Staphylococcus aureus, 4 Klebsiella pneumoniae, and 2 Pseudomonas aeruginosa. Following the primers design by the Primer3Plus software tool and optimization of the conventional polymerase chain reaction (PCR), the primers were validated for their use in the multiplex PCR experiments. The MFEprimer program was used to check the suitability of the primer set combinations for multiplex PCR. The MFEprimer software was successful in designing the multiplex-PCR experiments and de
