Extraction and Partial Purification of Exo-Polysaccharide from Lactobacillus Rhamnosus Isolated from Vaginal Specimens

Lactobacillus spp. is one of the most important strains used worldwide in different applications that range from medical to industrial uses. Therefore, this study aimed to determine the potential capability of the putative probiotic L. rhamnosus isolated from clinical vaginal specimens to produce exopolysaccharide (EPS). From a total of 100 clinical samples, only 13 (13%) samples were represented as Lactobacillus spp, as characterized by the use of the API 50CHL system. The results revealed that the number of L. rhamnosus isolates constituted 4/13 (30.8%), with a confident percentage of more than 80%. In addition, characterization by 16S rRNA sequencing showed 100% similarity to the characterized species of L. rhamnosus. Also, the results showed a strong capability of all Lactobacillus spp. isolates to produce biofilm. On the other hand, the antimicrobial susceptibility test revealed increases of antimicrobial resistance. The selected L. rhamnosus showed the
capability to produce exopolysaccharide with carbohydrate and protein concentrations of 74.774 μg/ml and 0.0145μg/ml, respectively. This bacterial isolate demonstrated the ability to form a thin capsule with carbohydrate and protein concentrations of about1.314 μg/ml and 0.01μg/ml, respectively.

Purification and Characterization β - lactamase produce from local isolate Klebsiella pneumonia

Beta-lactamase was purified from local isolate Klebsiella pneumonia by several steps included precipitation with ammonium sulphate at 20-40% saturation, DEAE- ion exchange chromatography and gel filtration on Sephacryl S-200 column. The obtained purification fold and recovery were 32.66; 47.04% respectively. The characterization of the purified beta-lactamase showed that the molecular weight was about 4000 daltons as determined by gel filtration.Purified enzyme had an optimal pH of 7 for activity and an optimal stability between pH 6.5-7.5, results shows that the optimal temperature appear to be 35 ? C .During storage the enzyme retained 72% at -20 ? C and retained 25% of the activity at the same period at 4 ? C.

Optimization of Pectinase Production from Pesudomonas sp. Isolated from Iraqi Soil

Identification and Purification of Cholera Like Toxin from Environmental Isolate of Vibrio cholerae

The presence and prevalence of V. cholerae were investigated in forty five water samples collected from different locations of Tiger River/ Baghdad city. Twenty one isolates were isolated by adopting a simple isolation techniques. The final identification revealed that only three isolates were confirmed as V. cholerae. Kinetics and Thermodynamics of Peppermint Oil Extraction from Peppermint Leaves

This study aimed to extraction of essential oil from peppermint leaves by using hydro distillation methods. In the peppermint oil extraction with hydro distillation method is studied the effect of the extraction temperature to the yield of peppermint oil. Besides it also studied the kinetics during the extraction process. Then, 2nd -order mechanism was adopted in the model of hydro distillation for estimation many parameters such as the initial extraction rate, capacity of extraction and the constant rat of extraction with various temperature. The same model was also used to estimate the activation energy. The results showed a spontaneous process, since the  Gibbs free energy had a value negative sign.

sulphate at 80 % saturation, DEAE- Cellulose Ion exchange chromatography and gel filteration on sepharose CL-6B column. The Final purification folds and the yield of the enzyme were 42.5 and 24.8 % respectively. The purified β-galactosidase has an optimum pH for its activity between 4.5 to 5, while the optimum pH for enzyme stability was between 5 to 5.5.
