Biofuel Production from Cellulosic Wastes by Local Isolates of Streptomyces sp.

The current study aims to produce cellulase enzyme from Streptomyces spp. isolates and study the effect of some cultural conditions on cellulase production; biofuel production from cellulotic waste through enzymatic and acids hydrolysis. Out of 74 isolates of Streptomyces sp. were screened for cellulse production in solid and liquid media. Results showed higher capability of isolate Streptomyces sp. B 167 for cellulase production and bioconversion of cellulose, therefore selected for further studies. The results of optimization revealed that the cellulase enzyme productivity by the selected isolate reached 2.1 and 2.28 U/ml after 48 h of incubation time and pH 7 respectively. Cellulase productions in tested isolate improved (2.57 U/ml) by supplementation of cellulose liquid medium with 1 % of yeast extract as nitrogen source. Additives of carbon sources like (manitol, glucose, maltose, sucrose and starch) to the process of saccharification not improve the cellulose productivity. The bioconversion of cellulosic waste to reducing sugar was maximum with Banana peels (77.78 %) followed by the rice husk (75.56 %), orange peels (71.11 %), corn steep peels (60.0 %) and lowest bioconversions (53.33 %) recorded with sawdust. The degradation of cellulosic waste increases with increasing substrate concentration. Maximum cellulase productivity (3.18 U/ml) and bioconversion (86.1 %) was obtained at 3 % (w/v) of cellulosic waste (Banana peels). Saccharification of cellulosic waste with different treatment methods was studied. The pretreatment of cellulosic waste with 1 % HCl and H2SO4 produces 21 and 15.8 g of reducing sugar / 100 g of cellulosic waste. In comparison, hydrolysis with Streptomyces sp. B 167 enzymes, resulting significantly higher amount of reducing sugar yield (25 g / 100 g cellulotic waste). Further fermentation of cellulosic hydrolysates were preformed using Saccharomyces cerevisiae using stationary fermentation condition, maximum yield of ethanol were ( 0.30, 0.19 and 0.10 g ethanol / g glucose) observed with Streptomyces sp. B 167 enzymes, HCl and H2SO4 hydrolysates respectively after 48 h of fermentation.

Genetic diversity of Capsicum annuum L. in local and imported samples in Iraq by using RAPD-PCR

Genetic variation was studied in 22 local and imported samples collected from local Iraqi market by using random amplified polymorphic DNA (RAPD-PCR). Five randomly primers set were used in this study. These primers produced 292 bands. Molecular weights of these bands ranged between 1.8 Kb (1800 bp) to 150 bp. The percentage of polymorphic bands is 100%, with one distinguished band which is produced by using C52 primer. The other primers did not produce any distinguished band. The results of Dendrogram of the studied samples depended on RAPD-PCR results by using Jaccard coefficient for genetic similarity was distributed the samples into 8 groups. This Dendrogram revealed a higher similarity between Iraqi/Yousifia green bell pepper and Jo

Production of Biogas from Mono- and Co-Digestion of Agricultural Waste (Cow Dung, Chicken Dropping, and Rice Husk)

This study focused on waste to energy technology that utilized mono- and co-digestion of cow dung (CD), chicken dropping (ChD), and rice husk (RH). The fabricated digesters were assessed for the influence of temperature and pH on biogas production from the materials used. The total aerobic bacteria and fungi counts for the mono- and co-digestion of cow dung with chicken droppings had highest number on day zero (1.5x107cfu/ml, 1.6x105cfu/ml and 1.4x108cfu/ml, 1.2x105, respectively), while the lowest counts were recorded on the 35th day (1.3x101cfu/ml, 1.0x101cfu/ml and 1.1x101cfu/ml, 1.0x101cfu/ml, respectively). The highest count of the ace

Molecular Detection of Suspected Leishmania Isolates Using Polymerase Chain Reaction

Leishmaniasis is a widespread parasitic disease that occurs as a result of infection with a unicellular parasite belonging to the genus Leishmania. Diagnosis by conventional methods is inaccurate and is not sensitive to confirm the genus infection. Here, we have investigated a methods for Leishmania genus diagnosis, which includes the technique of polymerase chain reaction to detect the presence of the parasite at in vitro for promastigote cultures using three genus-specific primer pairs to amplify HSP70, ITS, and ITS2. The results showed single band of ~1422, ~1020, and ~550 respectively. This study has proved the ability of these primer pairs to detect Leishmania infection and recommend them to be used for detection of leishmaniasis in

Detection of the Antiseptic Resistance Gene among Pseudomonas aeruginosa Isolates

Pseudomonas aeruginosa is an opportunistic pathogen that causes a number of infections in immunocompromised patients. This organism appears to improve resistance  to many antimicrobial agents and a high percentage of clinical isolates of P. aeruginosa exhibit multidrug resistance (MDR) phenotype . The purpose of this study is to screen the antibiotic susceptibility patterns and the prevalence of qacE delta1 gene among bacterial isolates. Accordingly, 145 samples were collected from different clinical sources from patients who admitted to different hospitals in Baghdad city in a period ranged 23/8/2018-1/1/2019. The isolates were diagnosed as P. aeruginosa based on routine b
