Induced callus from seedlings of Peganum harmala L. and studying harmine compound concentration in vitro and in vivo by GC analysis

Plant tissue culture considers a benefit biotechnological technique for scientific research especially the production of undifferentiation callus cells and regeneration through suspension or static media. The seedlings of Peganum harmala was used as a source to produce callus mass in vitro in static media through different tissue culture media supplemented by varying combinations of plant growth regulators (PGR). The result illustrates that 2 mg/l of Kinitine with 0.5 mg/l of 2, 4-D was efficient to stimulate callus induction with percent 100% in stem and root of P. harmala and this combination gave a high fresh weight, 1954 mg in root and 1170
mg in stem and high dry weight in root and stem was 74.60, 60.30 respectively. In a comparative analysis through gas chromatography (GC) the stem and root in field recorded harmine concentration 56.13 and 40.95 μg respectively, which was higher than the in vitro callus induction from stem and root, which may be due to the fact that field plants have not been exposed to plant hormones with concentrations higher than the normal level, which reduced the stimulation of cells producing active compounds.

The Effect of essential oil extracted from yellow peel of Citrus aurantium L. on growth of some fungi

  The study was conducted to evaluate the effect of essential oil extracted from yellow peels of Citrus aurantium on the radial growth of all fungi.  Penicillium expansum, Aspergilus flavus and Fusarium oxysporum.              The results showed significant gradual reduction of the surface growth of fungi, P. expansum and A. flavus was more affected by the essential oil, while  F. oxysporum  showed less sensitivity towards the essential oil. The reaction of growth was inconcommitant with increasing concentrationsof oil, reaching concentration of (5)% which showed complete inhibition.

Cytogenetic effect of crude aqueous extract from leaf of Lycium barbarum on root tip of Allium cepa L.

This study was carried out to investigate the cytogenetic effects of crude aqueous extract of Lycium barbarum on the roots tip of Allium cepa Using three concentration 125, 25, 50 mg/ ml for 2, 4, 6 hours treatment periods.This study were included some cytogenetic analysis such as mitotic index , phase index and chromosome aberration. The data showed that the treatment with 50mg/ml for 6huors led to reduce the mitotic index less than 50% . This reduction considered to have toxic and sublethal effect . These results revealed mutagenic potency by inducing differents type of chromosome aberration.

Abstract<p>The production of polyhydroxyalkanoates PHAs from biopolymer degrading bacteria was examined <italic>in situ</italic> by screening isolates using Sudan B Black staining process as potential PHAs detecting, and Nile Blue staining as a proof method detection. Materials and Methods: Ninety maxillary halves simulated fixed orthodontic appliances that were immersed in artificial saliva of different pH values (6.75, 5 and 3.5) during 28 day period.
