Fifty isolates of Bacillus sp. were subjected to the first and second screening to detect the ability to produce laccase enzyme and select the highest ones production of laccase on Petri plates containing nutrient agar supplemented with Cu2+.
Syringaldazine was used as an indicator and substrate for the determination of laccase activity. Three isolates, which consumed less time to developed pink color were tested for the production of laccase quantitatively. The effective isolate B16 with significant amounts of laccase 1.84 unit /ml was selected for laccase study.
The optimization studies revealed that the maximum laccase production was achieved when the production medium was at the following conditions: 5 days of incubation, temperature 35 C°, pH 7.0, copper sulphate 0.2 mM, galactose 3% as carbon source, 0.2% Tryptone as nitrogen source and K+ 1mM.

Two grades of paving asphalt with penetration of 46 and 65 are studied for determining changes in their physical and chemical properties caused by ageing.
The ageing process has been conducted on two petroleum paving asphalt cement using thin film oven test at 150, 163 and 175 C, and ageing time 5, 10,15, 20, 25 and 30 hours. The effect of ageing time and temperature on penetration, kinematic viscosity, softening point, solubility in trichloroethylene, heat loss and changes in chemical composition are investigated. Serum alkaline phosphatase activity has been investigated in three major types of tuberculosis (TB). These were Extra pulmonary primary, Acute miliary, and Primary tuberculosis. Activities were found elevated in these cases studied at basic PH and 37 C. The study concentrated an a comprehensive determination of the rate reaction kinetics of the enzyme reaction in both normal and TB sera. The pseudo first — order plot reflects both values of the first — order association constant (K I) and the half life time (t1/2) of the enzymatic reaction. The activation energy of the reaction (ES-complex formation) with Hill coefficient (n) were both estimated using Arrhenius and Hill plots respectively.
