Fifty isolates of Bacillus sp. were subjected to the first and second screening to detect the ability to produce laccase enzyme and select the highest ones production of laccase on Petri plates containing nutrient agar supplemented with Cu2+.
Syringaldazine was used as an indicator and substrate for the determination of laccase activity. Three isolates, which consumed less time to developed pink color were tested for the production of laccase quantitatively. The effective isolate B16 with significant amounts of laccase 1.84 unit /ml was selected for laccase study.
The optimization studies revealed that the maximum laccase production was achieved when the production medium was at the following conditions: 5 days of incubation, temperature 35 C°, pH 7.0, copper sulphate 0.2 mM, galactose 3% as carbon source, 0.2% Tryptone as nitrogen source and K+ 1mM.
Measurements of radon gas concentrations were carried out for 12 soil samples at 3 sampling depths (surface, 5 cm and 10 cm) collected from (4) locations in south Baghdad suburbs (Bu'aitha) using solid state nuclear track detector CR-39 and sealed can technique. Radon concentrations for surface samples were ranged from 402.2 to 1538.4 Bq.m-3 with an average 994.4 Bq.m-3. Whereas, radon concentration was ranged from 813.1to 2050.4 Bq.m-3 and from 1309.8 to 4626. 1Bq.m-3 with an average values of 1359.8 Bq.m-3 and 2338.3 Bq.m-3 for 5 cm and 10 cm depths respectively. A group of white mice were injected transurethrally using a catheter with S. saprophyticus S67 cell suspension in a concentration reached 109 CFU/ml. concomitantly, the role of its peptidoglycan in the pathogenicity was studied by injecting another group of mice with 0.3 mg/0.2 ml of partially purified S. saprophyticus S67 peptidoglycan extract. After autopsy, kidneys and urinary bladder showed several histopathological changes both in cells and peptidoglycan injected mice, included: hydropic degeneration, glomerulus shrinkage, congestion of renal vessels, infiltration of inflammatory cells, and dekeratinization in urinary bladder.
