Intra Cellular Fluorescence for Monitoring The Physiological Changes and Biostimulation of Yeast Irradiated with Low Power 532nm CW Nd:YAG Laser

The purpose of this study is to investigate the biostimulation effect of 532 nm CW laser on the metabolism of Saccharomyces cerevisiae yeast. Cells were irradiated by 532 nm Nd:YAG laser using 0.153 W/cm2 power density at 30, 45, 60,180 and 300 seconds exposure times in their respective orders. Intrafluorescence parameters were measured by detection the autofluorescence intensity, proliferation rate and Imaging the fluorescent mitochondria using confocal laser scanning microscope. The results showed that the 30 and 45 second exposure times seem to have stimulated changes in the cells that led to increase proliferation, viability and mitochondrial activity. Autofluorescence of cells increased after 45 and 60 seconds exposure time. After 300 seconds there seems to be very noticeable decrease in proliferation, viability and autofluorescence. Confocal microscopy images showed that here is a correlation between fluorescence intensity using mitochondrial probes and proliferation rates of cells.
Introduction
Saccharomyces cerevisiae is one of the most important fungi in the history of the world. This yeast is responsible for the production of alcoholic beverages and bread and a source of protein and was used in biotechnology and genetics as a host for the genes of other organisms (Madigan, et al. 2006). Low energy laser irradiation of which output power is in the range of mW modulates various biological effects and has been shown to have positive effect on living organisms both in vitro and in vivo. However, the true effect of low energy laser on cell proliferation is sill controversial, because of conflicting reports on the effects of visible laser light on the cells in culture (Antonio, et al., 2002).

Changes of the susceptibility of Staphylococcus aureus bacteria to the local therapeutic agent by using Nitrogen laser

This study involves the investigation of the effect of nitrogen laser with 337.1 nm wavelength on the sensitivity of Staphylococcus aureus bacteria by using local therapeutic due to burns. Thirty six isolate of Staphylococcus aureus bacteria were isolated from 25 patients suffering from sever burns, each isolate of bacteria was irradiated with nitrogen laser at (5, 10, 15 and 30) pulses/second repetition rates for 1, 5, 10, 20 and 30 minutes for each repetition rate. Selection of highly ethanol productive yeast

Seventy five isolates of Saccharomyces cerevisiae were identified, they were isolated from different local sources which included decayed fruits and vegetables, vinegar, fermented pasta, baker yeast and an alcohol factory. Identification of isolates was carried out by cultural microscopical and biochemical tests. Ethanol sensitivity of the isolates showed that the minimal inhibitory concentration of the isolate (Sy18) was 16% and Lethal concentration was 17%. Structural characterization of gamma irradiated ZnS thin films

The effects of gamma irradiation on the structure of ZnS films , which preparing by flash evaporation method, are studied using XRD. Two peaks of (111), (220) orientations are appeared in X ray chart indicating the cubic phase of the films .The lattice parameter, grain size, average internal stress, microstrain, dislocation density and degree of preferred orientation in the film are calculated and correlated with gamma irradiation.

FLUORESCENCE LIFETIME MEASUREMENT OF C5H6 AND C6D6

Absorption and lifetime measurements were made for C6H6. and C6D6 molecules in pure solid phase and in solid matrices of Ar, N2, CO, and CO2. All measurement are made at low temperatures ranging between 12-50 K. A modified least square fitting deconvolution was utilized to measure the fluorescence lifetime with an error less than 1%. The results show that the use of matrix isolation technique offer a unique environment that minimize the perturbation of the molecular or atomic system and hence facilitate spectral analysis.
