A spectrophotometric determination of azithromycin was optimized using the simplex model. The approach has been proven to be accurate and sensitive. The analyte has been reacted with bromothymol blue (BTB) to form a colored ion pair which has been extracted in chloroform in a buffer medium of pH=4 of potassium phthalate. The extracted colored product was assayed at 415 nm and exhibited a linear quantification range over (1 - 20) g/ml. The excipients did not exhibit any interferences with the proposed approach for assaying azithromycin in pharmaceutical formulations.
A simple analytical method was used in the present work for the simultaneous quantification of Ciprofloxacin and Isoniazid in pharmaceutical preparations. UV-Visible spectrophotometry has been applied to quantify these compounds in pure and mixture solutions using the first-order derivative method. The method depends on the first derivative spectrophotometry using zero-cross, peak to baseline, peak to peak and peak area measurements. Good linearity was shown in the concentration range of 2 to 24 µg∙mL-1 for Ciprofloxacin and 2 to 22 µg∙mL-1 for Isoniazid in the mixture, and the correlation coefficients were 0.9990 and 0.9989 respectively using peak area mode. A rapid and sensitive method for the determination of amoxicillin trihydrate (AMXT) based on the diazo-coupling reaction was studied. Sulphanilic acid diazotizes with nitrite ion in acidic medium to produce a water soluble, colorless diazonium ion, which subsequently coupled with AMXT to form a colored azo dye in the alkaline medium, having maximum absorption at 455 nm. The calibration graph showed that Beer's law is obeyed over the concentration range of 0.3 – 30.0 μg/mL of AMXT, with the detection limit of 0.15 μg/mL and molar absorptivity was 2.3 × 104 L/mol.cm. Method (B) is based on the quantitative quenching effect of terazosin on the native fluorescence of Eosin Y at the pH 3. The quenching of the fluorescence of Eosin Y was measured at 556 nm after excitation at 345 nm.
