Bioremediation of Imidacloprid by the Local Isolate Psychrobacter celer

Abstract<p>Pesticide biodegradation can be accomplished by the technique of bioremediation, which makes use of microorganisms’ ability to degrade pesticide residues. This study aimed to separate and identify imidacloprid-biodegradable from botanical fields soil of greenhouses in the Plant Protection Directorate /Ministry of Agriculture in Baghdad, which has been using imidacloprid pesticides for many years. Using high-performance liquid chromatography, residual imidacloprid concentrations in MSM medium at a concentration of 25 mg/L after 21 days were measured to identify the best degrading bacterial isolates. Isolate No.37 the best bacterial isolate was able to degrade 63% of imidacloprid. was identified as <italic>Psychrobacter celer</italic> according to morphological, cultural and biochemical tests. Moreover, genetic analysis for the 16S rRNA gene and given a new accession number (OP672320.1) in the GenBank of NCBI, from this study that the soil previously contaminated for long periods of time with imidacloprid can be treated by degrading the imidacloprid residues in it by isolated bacteria <italic>Psychrobacter celer.</italic> </p>

Determination of Optimal Conditions for the Production of Laccase Enzyme by Local Isolate of Bacillus sp.

Fifty isolates of Bacillus sp. were subjected to the first and second screening to detect the ability to produce laccase enzyme and select the highest ones production of laccase on Petri plates containing nutrient agar supplemented with Cu2+.
Syringaldazine was used as an indicator and substrate for the determination of laccase activity. Three isolates, which consumed less time to developed pink color were tested for the production of laccase quantitatively. The effective isolate B16 with significant amounts of laccase 1.84 unit /ml was selected for laccase study.

Optimization Kerosene Bio-degradation by a Local Soil Bacterium Isolate Klebsiella pneumoniae Sp. pneumonia

The active bacteria are isolated for kerosene degradation process. It has been found that Klebsiella pneumoniae sp. have the highest kerosene degradation which is 88.5%. The optimum conditions of kerosene degradation by Klebsiella pneumonia sp. are pH5, 48hr incubation period, 35°C temperature and 10000ppm the best kerosene concentration. Using local Iraqi Bacillus isolate to reduce water salinity

Microbial desalination cell (MDC) has been created for expelling water saltiness, power generation, and wastewater administration. The MDC comprised of three chambers (anode, center desalination, and cathode).Were tested ability of type locally isolated bacteria Bacillus spp.in produce electricity to water desalination. In recent study results showed that a remove where the salinity recorded 4000 ppm at room temperature at the voltages of 0.6 volts and less salinity at room temperature at 0.2 volts was 200 ppm. Recent results highlight the need to reduce time for reduce salinity decreased from 3500 ppm to 500 ppm the eleventh day at a voltage of 0.5 volts that depended on type of substrate.

Classical and Statistical Optimization of Medium Composition for Promoting Prodigiosin Produced by Local Isolate of Serratia Marcescens

Prodigiosin is a ‘natural red pigment produced by Serratia marcescens which exhibits immunosuppressive and anticancer properties in addition to antimicrobial activities. This work presents an attempt to maximize the production of prodigiosin by two different strategies: one factor at time (OFAT) and statistical optimization. The result of OFAT revealed that sucrose and peptone were the best carbon and nitrogen sources for pigment production with concentration of prodigiosin of about 135 mg/ L. Separation and Purification of Hemolysin from Local Isolate of Serratia marcescens

Twenty isolates of Serratia marcescens were isolated from inflammation of the urinary tract (UTI)., These isolates were found to produce hemolysin as indicated by blood agar plates in which the hemolysis of red blood cell indicate a positive result. Isolates were selected according to their hemolysis activity by measuring absorbance of hemoglobin at 405 nm that released from red blood cell. Hemolysin was completely purified using 50-75% saturation of ammonium sulphate followed by ion exchange chromatography with DEAE-cellulose then gel filtration chromatography by sepharose 4B. Accordingly molecular weight for the purified toxin was estimated as 45 KD.
