A new series of cyclic coumarin-fluorocytosine hybrid conjugates (N1-N6) was synthesized through microwave technology. The synthetic plan involved the formation of the required Schiff base N, followed by the transformation of the azomethine functional group of the Schiff base N to oxazepine and imidazolidone rings. The antimicrobial activity of the synthesized compounds N-N6 was assessed against Gram-positive bacteria (Staphylococcus aureus and Staphylococcus epidermidis) and Gram-negative bacteria (Escherichia coli, Pseudomonas, and Klebsiella) using the disk diffusion method. Compounds N4 and N5 showed the highest activity against S. aureus (24 mm inhibition), with N5 also demonstrating superior inhibition against Klebsiella and Pseudomonas. Molecular docking studies with DNA gyrase enzymes (Protein Data Bank: 1AJ6 for (E. coli) and 3G7B for (S. aureus) revealed that compound N5 strongly binds to both (E. coli) (–9.28 kcal/mol) and (S. aureus) (–9.65 kcal/mol), involving hydrogen bonds, hydrophobic interactions, and halogenation at conserved active site residues. All compounds met Lipinski's criteria, did not cross the blood-brain barrier, and displayed favorable gastrointestinal absorption and bioavailability according to absorption, distribution, metabolism, and excretion predictions. Overall, these hybrid compounds, especially N5, show promise as antimicrobial agents.. KEYWORDS :Coumarin, Fluorocytosine, Cycloaddition, Molecular docking.
Schiff bases (Sh1-Sh3) have been synthesized (p-aminophenol) was condensed with different aromatic aldehyde in ethanol inthe presence of glacial acetic acid as catalyst. These Schiff bases on treatment with monochloroacetyl choride gave 3-chloro-1-(4-hydroxyphenyl)-4-(substituted)azetidin-2-one(Az4-Az6), with αmercaptoacetic acid gave 3-(4-hydroxyphenyl)-2-( substituted)thiazolidin-4-one (Th7-Th9) and with anthranilic acid gave 3-(4-hydroxyphenyl)-2(substituted)-2,3-dihydroquinazolin-4(1H)-one (Qu10-Qu12). The purity of the derivatives was confirmed by TLC. The some compoundsidentify by (FT-IR and1H, 13C-NMR) data. Show MoreWith the aim of developing potential antimicrobials, a series of novel Ciprofloxacin methylene isatin derivatives incorporating different aromatic aldehydes were synthesized and characterized by FTIR, 1H NMR, Mass spectroscopy and bases of elemental analysis. In addition, the in vitro antibacterial and antifungal properties were tested against some human pathogenic microorganisms by employing the disc diffusion technique. A majority of compounds were showing activity against several of the microorganisms. The relationship between the functional group variation and the biological activity of the evaluated compounds is discussed. From comparisons of the compounds, 3c was determined to be the most active compound.
Objective:This study involved synthesis of a new series of different five-membered heterocyclic derivatives, testing their antioxidant activity, and examining their potential in vitro antimicrobial agents. Methods: The synthesis of the derivatives involved a three-step process. Initially, succinyl chloride was reacted with methanol, followed by a reaction with 80% hydrazine hydrate through a nucleophilic addition-elimination mechanism, resulting in the formation of succinohydrazide (I). This compound was then employed as a precursor for the synthesis of Schiff bases (II), and (III) by reacting it with m-nitro benzaldehyde and p-nitro benzaldehyde. Following this, a ring closure reaction was applied using thioglycolic acid, glycolic acid,
