Transcriptomic Reanalysis Reveals Complement, Circadian, and Toll-Like Receptor Signaling Remodeling in Lactiplantibacillus plantarum-Induced Innate Immune Memory in Human Monocytes
...Show More Authors

The commensal bacterium Lactiplantibacillus plantarum is recognized for its ability to cause a state of innate immune memory in monocytes, characterized by the tolerogenic phenotype and lowered pro-inflammatory responses. This study provides an extensive re-examination of the transcriptomic dataset originating from human CD14 + monocytes primed with live or heat-killed L. plantarum (GSE159496). Using a computational approach, 1,954 differentially expressed gene calls that corresponded to 1,397 unique genes were obtained through three pairwise comparisons. In addition, several novel biological pathways that were not investigated in the original study were discovered. Priming with live L. plantarum resulted in a marked downregulation of the complement system (12.5% decrease in its module score, p < 0.001), indicating an important mechanism of immune evasion. Additionally, a significant alteration of the circadian rhythm pathway was revealed, indicating its connection to metabolic time management in sustaining long-term immune tolerance. Toll-like receptor (TLR) signaling was coordinately repressed, with 17 pathway genes—including the receptors TLR2 and TLR8—significantly downregulated in live primed cells (p.adjust = 3.0 × 10–6 ). Other significantly modulated novel pathways included a downregulation of glycosylation and post-translational modifications (−13.2%, p < 0.01) and an upregulation of amino acid sensing/mTOR signaling (+11%, p < 0.001) and xenobiotic detoxification (+12%, p < 0.01). These findings point to a multi-layered adaptive response where L. plantarum not only dampens canonical inflammatory signaling but also actively remodels cellular processes related to pathogen recognition, metabolic sensing, and stress responses. This reanalysis provides a more granular model of probiotic-induced immune memory. Received: 24 June 2026 | Revised: 6 August 2026 | Accepted: 15 September 2026 Conflicts of Interest The authors declare that they have no conflicts of interest to this work. Data Availability Statement The data analyzed in this study are publicly available in the NCBI Gene Expression Omnibus under accession GSE159496: https://www.ncbi.nlm.nih.gov/geo/query/acc.cgi?acc=GSE159496. Synergistic effects of NH2-MIL-101(Fe) metal-organic framework and Pd nanoparticles for sensitive determination of norepinephrine in the presence of acetaminophen
...Show More Authors

An electrochemical sensor based on an amino-functionalized iron NH2-MIL-101(Fe) metal-organic framework (MOF)/Pd nanoparticles (NPs) composite-modified screen-printed electrode (SPE) is prepared for the simultaneous determination of norepinephrine (NEPI) and acetaminophen (ACP). The NH2-MIL-101(Fe) MOF/Pd NPs/SPE electrochemical sensor shows a significant enhancement in the response peak current of NEPI, as compared to bare SPE. This suggests that the unique features of NH2-MIL-101(Fe) MOF/Pd NPs composite-modified SPE improve the electrocatalytic oxidation of NEPI. Such a synergistic effect between NH2-MIL-101(Fe) MOF and Pd NPs results in a significant enhancement in the response, where the MOF's high surface area co

Genotyping of IL-4 −590 (C>T) Gene in Iraqi Asthma Patients
...Show More Authors

This study is the first investigation in Iraq dealing with genotyping of IL-4 −590 (C>T) gene, especially in Iraqi patients with asthma. We studied forty-eight blood samples collected from patients with asthma and compared with age-matched 25 healthy individuals as controls. The polymorphism results of IL-4 −590 (C>T) gene by using amplification refractory mutation system (ARMS-PCR) showed the presence of C and T alleles and three genotypes (CC, CT, and TT). Interestingly the frequency of C allele and CC genotype was higher in patients with asthma in comparison with the same allele and geno

Toll like receptor 2(TLR2) in acne vulgaris patients before and after oral isotretinion therapy
...Show More Authors

Background: Acne vulgaris is one of the top three most commonly encountered dermatological problems worldwide in both primary and secondary care. Human keratinocytes express functional TLR2 heterodimers. An increased expression of TLR2 was detected in the epidermis of inflammatory acne lesions, as observed in normal skin; the expression level increased with the degree of differentiation of the keratinocytes. TLR2 expression is upregulated in inflammatory acne lesions and induced by C. acnes. The current study conducted to assess the oral isotretinion treatment effect on the acne vulgaris patients by evaluated the Toll Like Receptor 2 as a major immune system marker in Acne vulgaris immune re

By March 2020, a pandemic had been emerged Corona Virus Infection in 2019 (COVID-19), which was triggered through the sensitive pulmonary syndrome (SARS disease corona virus- 2 (SARS COV-2). Overall precise path physiology of SARS COV-2 still unknown, as does the involvement of every element of the acute or adaptable immunity systems. Additionally, evidence from additional corona virus groups, including SARS COV as well as the Middle East pulmonary disease, besides that, fresh discoveries might help researchers fully comprehend SARS CoV-2. Toll-like receptors (TLRs) serve a critical part in both detection of viral particles as well as the stimulation of the body's immune response. When TLR systems are activated, pro-inflammatory cy
