Transcriptomic Reanalysis Reveals Complement, Circadian, and Toll-Like Receptor Signaling Remodeling in Lactiplantibacillus plantarum-Induced Innate Immune Memory in Human Monocytes
...Show More Authors

The commensal bacterium Lactiplantibacillus plantarum is recognized for its ability to cause a state of innate immune memory in monocytes, characterized by the tolerogenic phenotype and lowered pro-inflammatory responses. This study provides an extensive re-examination of the transcriptomic dataset originating from human CD14 + monocytes primed with live or heat-killed L. plantarum (GSE159496). Using a computational approach, 1,954 differentially expressed gene calls that corresponded to 1,397 unique genes were obtained through three pairwise comparisons. In addition, several novel biological pathways that were not investigated in the original study were discovered. Priming with live L. plantarum resulted in a marked downregulation of the complement system (12.5% decrease in its module score, p < 0.001), indicating an important mechanism of immune evasion. Additionally, a significant alteration of the circadian rhythm pathway was revealed, indicating its connection to metabolic time management in sustaining long-term immune tolerance. Toll-like receptor (TLR) signaling was coordinately repressed, with 17 pathway genes—including the receptors TLR2 and TLR8—significantly downregulated in live primed cells (p.adjust = 3.0 × 10–6 ). Other significantly modulated novel pathways included a downregulation of glycosylation and post-translational modifications (−13.2%, p < 0.01) and an upregulation of amino acid sensing/mTOR signaling (+11%, p < 0.001) and xenobiotic detoxification (+12%, p < 0.01). These findings point to a multi-layered adaptive response where L. plantarum not only dampens canonical inflammatory signaling but also actively remodels cellular processes related to pathogen recognition, metabolic sensing, and stress responses. This reanalysis provides a more granular model of probiotic-induced immune memory. Received: 24 June 2026 | Revised: 6 August 2026 | Accepted: 15 September 2026 Conflicts of Interest The authors declare that they have no conflicts of interest to this work. Data Availability Statement The data analyzed in this study are publicly available in the NCBI Gene Expression Omnibus under accession GSE159496: https://www.ncbi.nlm.nih.gov/geo/query/acc.cgi?acc=GSE159496. Asthma is chronic inflammatory disease affecting 5% of world population. Characterized by eosinophilic type2 inflammation. FKBP51 immunophilin, important modular protein of glucocorticoid receptor (GR). We aimed to evaluate immunocytochemical localization of GR and FKBP51 in induced sputum cells by using immunocytochemical method and immunofluorescent ant-FKBP51 and anti –GR antibody and estimation of IgE and Type 2 inflammatory cytokine IL-5,IL-13 by ELISA technique.GR in the sputum show non-significant decrease of cytoplasmic distribution of the patient groups and highly significant increase in steroid treated patients and non-significant increase in nuclear distribution in non-steroid, FKBP51 nuclear localization show non-significant i

Effects of Hydrochlorothiazide on Tenofovir Disoproxil Fumarate-Induced Nephrotoxicity in Rats

Tenofovir disoproxil fumarate, a nucleotide reverse transcriptase inhibitor utilized for the treatment of hepatitis B virus and human immunodeficiency virus infections; and is now one of the most widely used antiretroviral drug. However, tenofovir disoproxil fumarate can induce nephrotoxicity, which may be attributed to the interaction between such drug and the organic anion transporters (hOAT1, and OAT3) with consequent changes in levels of some parameters that may have a role in nephrotoxicity. Thiazide diuretics have high to intermediate potency of inhibition of OAT1s and OAT3; thus, it may possess nephroprotective effects. The Morphological and Histopathological Liver Abnormalities Caused by Carbamazepine-Induced Injury in Female Albino Mice

Background: The adverse effects of drugs can damage various organs, especially the liver, leading to a hepatic injury known as hepatotoxicity. Drug-induced liver injury (DILI) is challenging nowadays because of the large number of different drugs used, one of the offending medications that cause DILI is carbamazepine (CBZ), since the liver has an array of functions including detoxification, it will deal with several damages caused by exposure to the drugs. Objective: investigate the effect of (CBZ) 20mg/kg/day on female mice liver after 14 and 30 days of treatment on morphological and histopathological levels. Materials and Methods: 20mg/kg/day of CBZ was administered orally for (14) days to (10) female mice, another (10) mice were taking t

Influence of working pressure and lasing energy of Al plasma in laser-induced breakdown spectroscopy
...Show More Authors

Aluminum plasma was generated by the irradiation of the target
with Nd: YAG laser operated at a wavelength of 1064 nm. The
effect of laser power density and the working pressure on spectral
lines generating by laser ablation, were detected by using optical
spectroscopy. The electron density was measured using the Stark
broadening of aluminum lines and the electron temperature by
Boltzmann plot method it is one of the methods that are used. The
electron temperature Te, electron density ne, plasma frequency
and Debye length increased with increasing the laser peak
power. The electron temperature decrease with increasing gas
pressure.
