Extraction and Purification of Indole aectic acid from locale isolate Fusarium oxysporum(F2)

Indole acetic acid (IAA) produced from F. oxysporum (F2) was purified by several steps included extraction by cold ethyl acetate ; Column chromatography using silica gel and TLC chromatography . The pure indole acetic acid (IAA) which produce by F. oxysporum (IAA) was tested by ultraviolet spectra at (200-300)nm ; and appear that the maximum absorbance at 229nm , the high performance liquid chromatography (HPLC) used to test the purity of the indole acetic acid and the results showed one peak at appearance time 3.822 min

Classical and Statistical Optimization of Medium Composition for Promoting Prodigiosin Produced by Local Isolate of Serratia Marcescens

Prodigiosin is a ‘natural red pigment produced by Serratia marcescens which exhibits immunosuppressive and anticancer properties in addition to antimicrobial activities. This work presents an attempt to maximize the production of prodigiosin by two different strategies: one factor at time (OFAT) and statistical optimization. The result of OFAT revealed that sucrose and peptone were the best carbon and nitrogen sources for pigment production with concentration of prodigiosin of about 135 mg/ L. This value was increased to 331.6mg/ L with an optimized ratio of C/N (60:40) and reached 356.8 with pH 6 and 2% inoculum size at end of classical optimization. A series of new metal complexes of this ligand were prepared by a reaction with the chlorides of Cd(II), Co(II), Ni(II), Cu(II) and Zn(II). The research includes preparation of a new Schiff base from the condensation of sulfonic acid with 2-phthalaldehyde by dissolving (0.2 g, 0.346 mmole) of the ligand (L) in (10 ml) of absolute ethanol, then 0.06 g of potassium hydroxide (KOH) dissolved in 10 ml of distilled water added to it and reacting the product with some binary metal salts (Mn, Co, Ni, Cu, Cd, Zn, Pt, Hg).
