This study focuses on diagnosis of Candida species causing Vulvovaginal Candidiasis using phenotype and genotype analyzing methods, and frequencies of candida species also using Vulvovaginal Candidiasis patients. 130 samples (100 from patients and 30 from non infected women) were collected and cultured on biological media. Identifying the yeasts, initially some phenotypic experiments were carried out such as germ tube, from motion of pseudohyphae and clamydospores in CMA+TW80 medium, API20 candida and CHROMagar Candida. Genomic DNA of all species were extracted and analyzed with PCR and subsequent Polymerase Chain Reaction - Restriction Fragments Length Polymorphism (PCR-RFLP) methods. Frequency of C. albicans, C. krusei, C. tropicalis , C. parapsilosis and C. glabrata were 46.4%, 31%, 18%, 7.2%, and 1.8%, respectively.The ITS1-ITS4 region was amplified and the Restriction enzyme Msp1 digests this region and was used to identify of candida species .Electrophoretically ribosomal DNA of C. albicans, C. krusei, C. tropicalis and C. glabrata produced two bands whereas the C. parapsilosis gave one band.
Two hundred staphylococcal isolates isolated from milk and white cheese samples, which were collected from local markets in Baghdad. The predominant species was Staphylococcus aureus 97 isolates (48.5%), followed by S.chromogenes 82 (41%) and 21 (10.5%) S.epidermidis isolates. The pattern of antibiotic susceptibility of Coagulase Positive Staphylococci (COPS) and Coagulase Negative Staphylococci (CONS) isolates to 3 antibiotics (Methicillin, Tetracyclin and Vancomycin) was determined using disc diffusion method; the results revealed that 80 S. aureus isolates (82.47%) found to be methicillin resistant (MRSA) while 8 isolates (8.24%) were vancomycin resistant (VRSA) and 18 S. aureus isolates (18.5%) resist tetracycline antibiotic. Amoebas live freely in different climates and parts of the world. Several species of Free Living Amoeba (FLA) are capable of causing serious as well as fatal infections in human beings. The aim of this study was to identify and compare genotypes of water-polluting FLA in major rivers and lakes of Iraq and compare them with FLA isolates from Iran and Turkey. For this purpose, the study included 20 water samples from the Tigris River, Euphrates River , Najaf Sea and Dukan lake in Iraq, 20 water samples from Marivan, Velasht, and Soleimanshah lakes and Caspian sea in Iran, and 20 water samples from Sabanca, Seyfi , Hazar and Yay lakes in Turkey. The current study was done to record the infection rate of cryptosporidiosis in human and cattle, and genotype the clinical isolates of Cryptosporidium in Baghdad Province. A total of 265 stool sample were collected (150 from human and 115 from cattle) during the period from December 2016 to the May 2017. Cryptosporidial infection was detected using modified acid fast stain. DNA of the parasite was extracted from oocysts of positive fecal samples and nested PCR method was used for partial 60 kDa glycoprotein (gp60) gene amplification then sequence analysis for selected samples.
