Preparation, Characterization, and in - vivo Evaluation of Daptomycin Poly(D,L-lactide-co-glycolide) Microspheres

             Polymeric microsphere devices occupy a wide range in the field of controlled drug delivery. Subcutaneous injectable preparations of Poly(Lactide-co-Glycolide) (PLGA) microsphere of Daptomycine were prepared by solvent extraction/evaporation technique using different copolymers ratio and molecular weights. Four formulations were prepared (F1-F4) and characterized in term of particle size, surface morphology, bulk density and porosity in addition to the drug content. The effects of the above parameters on the in-vitro release study were evaluated. These formulas were evaluated also for their in-vivo release profile using rat (as an animal model) and their serum daptomycin concentration were evaluated accordingly. A trapezoidal method was used to estimate the cumulative AUC parameter and the effect of copolymers ratio and molecular weights on the shape of AUC for four formulas were evaluated. Simulation study was performed (for 7 or 10 days dosing of F1 and F2 and 15 day dosing for F3 and F4) as a predictor of the steady state concentration and other pharmacokinetics parameters. It was inferred from this study that by using tailored formulation approach, long and short acting preparations of injectable daptomycin microsphere could be developed helping to provide the clinicians more flexibility to select the suitable preparation according to the patient need.

Cytogenetic effects of insecticide Lamda cyhalothrin on meristemic cells of Allium cepa L.

The effect of insecticide lamda cyhalothrin on Allium cepa L.root meristem were studied cytogenetically .Using three concentrations of the insecticide 5%, 7.5%, 10% at 6,12,24hours treatments . The insecticide significantly reduced the mitotic index at all concentrations. Moreover showed its ability to induced C – metaphase in 10% .The effects were also caused chromosome aberration like stickness in anaphase, telophase, binucleated cells, Micronuclei formation. These alternations indicating that this insecticide was effective in producing disturbance of spindle fibers.

Purification and Characterization of Endoglucanase from local isolate of Aspergillus flavus

Endoglucanase produced from Aspergillus flavus was purified by several steps including precipitation with 25 % ammonium sulphate followed by Ion –exchange chromatography, the obtained specific activity was 377.35 U/ mg protein, with a yield of 51.32 % .This step was followed by gel filtration chromatography (Sepharose -6B), when a value of specific activity was 400 U/ mg protein, with a yield of 48 %. Certain properties of this purified enzyme were investigated, the optimum pH of activity was 7 and the pH of its stability was 4.5, while the temperature stability was 40 °C for 60 min. The enzyme retained 100% of its original activity after incubation at 40 °C for 60 min; the optimum temperature for enzyme activity was 40 °C.

Preparation of samarium doped-PMMA composite by casting method to evaluate the optical properties and potential applications

Samarium ions (Sm +3), a rare-earth element, have a significant optical emission within the visible spectrum. PMMA samples, mixed with different ratios of SmCl3.6H2O, were prepared via the casting method. The composite was tested using UV-visible, photoluminescence and thermogravimetric analysis (TGA). The FTIR spectrometry of PMMA samples showed some changes, including variation in band intensity, location, and width. Mixed with samarium decreases the intensity of the CO and CH2 stretching bands and band position. A new band appeared corresponding to ionic bonds between samarium cations with negative branches in the polymer.
