Cytotoxic effect of Gliotoxin from Candida spp. isolated from clinical sources against cancer and normal cell lines

Background: Invasive fungal infections have become more common during the past two decades. Candida species are the most common human fungal infections. Internal injuries characterize these infections because of virulence factors, such as gliotoxin, which is a fungal toxin that is thought to be antibacterial, antifungal, and antiviral.

Objectives: To test the ability of Candida species obtained from clinical sources to produce gliotoxin as a virulence factor and investigate its cytotoxicity effects against some selected cell lines.

Materials and Methods: One hundred and ten clinical isolates of Candida species were obtained from patients attendi

Cytotoxic effects of CeO2 NPs and β-Carotene and their ability to induce apoptosis in human breast normal and cancer cell lines

Cerium Oxide Nanoparticles (CeO2 NPs) and β-carotene are natural-source products that have recently gained an increased interest as pharmaceutical additives because of their effectiveness in living systems, but the behavior of these substances varies according to factors and conditions. The above mentioned materials were evaluated in breast normal (HBL-100) and cancer cell lines (CAL-51 and MCF-7 ) by different techniques ; MTT assay for studying cytotoxic effects, morphological changes, sqPCR, including gene expression of caspases 8 and 9, and P53. All experiments were conducted on cell lines by the use of the materials alone as well as their combination.

Synthesis and Characterization of Nanoparticles Extracted from Catharanthus roseus Plant

The Catharanthus roseus plant was extracted and converted to nanoparticles in this work. The Soxhlet method extracted alkaloid compounds from the plant Catharanthus roseus and converted them to the nanoscale. Chitosan polymer was used as a linking material and converted to Chitosan nanoparticles using Sodium TriPolyPhosphate (STPP). The extracted alkaloids were linked with Chitosan nanoparticles CSNPs by maleic anhydride to get the final product (CSNPs- Linker- alkaloids). The synthesized (CSNPs- Linker- alkaloids) was characterized using SEM spectroscopy UV–Vis., Zeta Potential, and HPLC High-Performance Liquid Chromatography. Scanning electron microscope (SEM) analysis shows that the Chitosan nanoparticles (CSNPs) have small dim

Expression of Recombinant Human Glucocerebrosidase Protein in Sunflowers

Molecular farming has become one of the most significant implementations of modern biotechnology to generate modified plant crops to produce medicinal proteins. Agrobacterium is one plant genetic engineering tool that integrates genes of interest inside a host plant. In recent years, the need to produce recombinant proteins as therapeutics has growing rapidly, and human glucocerebrosidase is one of the proteins that is need to treat disease. In this study, specific primers were designed to amplify Hu-GBA1 gene from constructed pGEM-GBA plasmid which was cloned into the plant expression vector pCAMBIA1304. The generated recombinant pCAMBIA1304-GBA plasmid was used to transform A. tumefaciens LBA4404

Apoptotic Activity of methionine γ- lyase on several cancer cell lines

The apoptotic activity of methionine γ- lyase from Pseudomonas putida on cancer cell lines was indicated by measuring the concentration of cytochrome c in the supernatants of cell lines. The result revealed high concentration of cytochrome c in the supernatants of cancer cell lines (RD, AMGM and AMN3) respectively while the concentration of anti-apoptotic protein (Bcl-2) was very low.

P53 mRNA in- Situ Hybridization analysis and Immunohistochemical Expression in Lung Cancer: A Comparative Study.

Background and objectives: P53 gene mutation and deletion are among the important molecular markers linked to lung cancer. In most cases, the inactivating mutations affecting both p53 alleles are acquired in somatic cells. Less commonly, the mutations are inherited ones. The aim of the present study was to analyze the frequency of having a wild and/or a mutated p53 gene in lung cancer compared to benign lung lesions and to relate these findings to different morphological types and grades of lung cancer.
Patients, materials and methods: In this retrospective study, the histopathology blocks of 30 lung cancer cases covering the period from2002 to 2007were obtained from the archives of the histopathology sec

High Expression of P53 protein in Recurrent Pregnancy Loss Could Play a Role in the Pathology!!

Background: Recent data provide evidence that p53 plays a critical role in mediating pregnancy by regulating steroid hormone activation
Objective: localization of semi quantitation tof por protein at the materno-fetal interface, in patients with recurrent pregnancy loss.
Methods: Immunohistochemistry analysis of p53 protein using paraffin embedded sections of curate samples obtained from 40 women, who where divided into three groups: 24 women with RPL, 10 women with abortion for the first time, and 6 women with induced abortion.
Results: The mean value of the expression of p53 protein in the RPL group was (65.8±2.16), which is significantly higher than that of the second group (48.0±2.81), and th

Reliable Reference Gene for Normalization of RT- qPCR Data in Human Cancer Cell Lines
Subjected to Gene Knockdown

Quantitative real-time Polymerase Chain Reaction (RT-qPCR) has become a valuable molecular technique in biomedical research. The selection of suitable endogenous reference genes is necessary for normalization of target gene expression in RT-qPCR experiments. The aim of this study was to determine the suitability of each 18S rRNA and ACTB as internal control genes for normalization of RT-qPCR data in some human cell lines transfected with small interfering RNA (siRNA). Four cancer cell lines including MCF-7, T47D, MDA-MB-231 and Hela cells along with HEK293 representing an embryonic cell line were depleted of E2F6 using siRNA specific for E2F6 compared to negative control cells, which were transfected with siRNA not specific for any gene. Evaluating the in vitro Cytotoxicity of Thymus vulgaris Essential Oil on MCF-7 and HeLa Cancer Cell Lines

Thyme essential oil (TEO) was extracted from dried leaves of Thymus vulgaris. The air-dried aerial parts of the plant produced 1.0% yield of TEO. The detection of this essential oil’s compounds was performed by GC-MASS. The cytotoxic activity of TEO was evaluated against two human cancer cell lines, namely HeLa (human epithelial cervical cancer) and MCF-7 (human breast carcinoma). Cells grown in 96 multi-well plates were treated with six concentrations of EO (6.25, 12.5, 25, 50, 100, 200 ppm) and incubated at 37 °C for 72 hrs. Cancer cell lines elicited various degrees of sensitivity to the cytotoxic effect of essential oil. The TEO exhibited significant differences (p≤ 0.01) between the effects of

The Pharmacological Effects of Kappa Carrageenan on Different Human Cell Lines and Genomic DNA: An in vitro study

Carrageenan extract is a compound of sulfated polyglycan that is taken out from red seaweeds. Being hydrocolloid in nature, carrageenan has gelling, emulsifying and thickening properties allowing it to be commonly used in the oral healthcare products and cosmetics. Due to its bioactive compounds, carrageenan has been shown to have antimicrobial, antiviral, and antitumor properties. The purpose of this work is to study the probable use of carrageenan on the diseases that are related to oral cavity and on the genomic DNA in in vitro experimental model

In this study, the effects of k-carrageenan on four different cell lines related to the cancer and normal cells which cultured on selective media were done. Moreover, the eff
