    A Simple, rapid and sensitive extractive and spectrophotometric method has been described for the analysis of diphenhyldramine –HCl (DPH) in pure form and in pharmaceutical formulations. The method is based on the formation of chloroform soluble ion-pair  complex with Bromophenol blue(BPB) in a phthalate buffer at pH 3.0.The extracted complex shows maximum absorbance at 410 nm. Beer's law is obeyed in the concentration range 0.2-25.0 µg.ml-1. The molar absorptivity and Sandell's sensitivity for the system being  2.416x104 L.mol-1.cm-1 and 0.012µg.cm-2, respectively.      Five hundred nasal swabs were taken from normal medical staff and public in the city of Baghdad. Several identification parameters were used to recognize the bacterial isolates.  S. aureus isolations form nasal swabs were identified using morphology and VITECK 2 system. Polymerase chain reaction (PCR) was employed to determine PVL (Panton–Valentine leukocidin ) gene in S. aureus. The data showed no significant evidence on the relationship between PVL gene presence and gender and age of  the studied groups. There was no relation between the prevalence of PVL gene in the age groups  of  21-30 years (p=0.328) and 31-40 years (p=0.682).

Ninety nine swabs were collected from patients with diabetic foot ulcers (DFU), all swabs   were cultured on different selective media for screening, 46 isolates confirmed as S. aureus by API staph. The results of antibiotic susceptibility test revealed that all isolates were resistant to metronidazole, 34 isolates were resistant to cefoxitin, ceftriaxone, and meropenim, 23 isolates were resistant to ciprofloxacin and norfloxacin, 17 and 16 isolates were resistant to tetracycline and trimethoprim, respectively; while all isolates were sensitive to tigecycline. Out of 150 clinical samples, 50 isolates of Klebsiella pneumoniae were identified according to morphological and biochemical properties.
