The protective role of nicardipine in dextran sulfate sodium-induced colitis in mice: Modulating inflammation and apoptosis
...Show More Authors

Ulcerative colitis (UC) is a chronic inflammatory bowel disease associated with persistent inflammation, oxidative stress, and epithelial apoptosis. Nicardipine, a dihydropyridine calcium channel blocker, exhibits anti-inflammatory and anti-apoptotic properties, but its therapeutic potential in UC remains unclear. This study evaluated the effects of nicardipine on dextran sulfate sodium (DSS)-induced colitis in mice, focusing on inflammatory, oxidative, and apoptotic pathways. Fifty BALB/c mice were assigned to five groups (n = 10): control, DSS, nicardipine 12 mg/kg, nicardipine 24 mg/kg, and 5-aminosalicylate (ASA) 75 mg/kg. Treatments were administered for 3 days before and 10 days during DSS exposure. Disease severity was assessed by body weight, disease activity index (DAI), and colon length. Colonic mRNA levels of Nlrp3, TNF-α, IL-17, and TNFSF10 were quantified by RT-PCR; protein expression of caspase-3, caspase-8, BAX, and BCL-2 was analyzed by Western blot. Serum malondialdehyde (MDA), myeloperoxidase (MPO), glutathione peroxidase-1 (GPX-1), occludin, and prostaglandin E₂ (PGE-2) were measured by ELISA. Histological scoring assessed epithelial integrity and inflammation. Nicardipine dose-dependently reduced DSS-induced weight loss, DAI, and colon shortening. Both doses significantly downregulated Nlrp3, TNF-α, IL-17, and TNFSF10 (p < 0.05), decreased caspase-3 and BAX, and increased BCL-2. Nicardipine restored GPX-1, lowered MDA and MPO, preserved occludin, and reduced PGE-2. Histology confirmed reduced mucosal injury and preserved epithelial architecture. Nicardipine attenuates DSS-induced colitis by suppressing pro-inflammatory cytokines, reducing oxidative stress, and inhibiting apoptosis, supporting its potential as a therapeutic candidate for UC. Further studies are warranted to clarify its molecular mechanisms and clinical relevance.

The Role of Lipopolysaccharide (LPS) in Modulating the Immune Reaction after Infected with Leishmania Donovani in Albino Mice
...Show More Authors

Aim: The aim of this study was to investigate the effect of LPS in immune response through detecting some immune markers by IHC technique in the liver of mice infected with Leishmania donovani (VL). Methods: Three groups of eighteen mice were created: the first was control negative (non-infected), the second was control positive (infected with 2X107 promastigotes of Leishmania donovani), and the third group was infected with 2X107 promastigotes of Leishmania donovani and treated with 40 ng/ml of LPS. The treatment was given orally twice daily for one month. Mice were dissected, and the liver separated for an immunohistochemistry study for the markers (BCL-2, Caspase-3, CD3, CTLA-4). Results: The results showed there was significant elevatio

The Protective Effect of Ethanolic Extract of Mentha spicata Against Irinotecan-Induced Mucositis in mice
...Show More Authors

Irinotecan induced-mucositis is an inflammatory event of intestine caused by an increase in concentration of active metabolite 7ethyl10-hydroxycamptothecin (SN38) in the intestine. Irinotecan must first be converted by a carboxylesterase (CES) to the active metabolite (SN38), which is subsequently glucuronidated by the hepatic enzyme to SN38G. The SN-38G is deconjugated in the intestine to SN-38 via ?-glucuronidase produced by the intestinal bacterial flora, which accounts for SN-38 delayed intestinal mucositis of irinotecan. To study the protective effect of mentha in irinotecan-induced mucositis, intestinal mucositis induced by I.P injection of irinotecan (75mg/Kg/day) for 4 days. Mentha ethanolic extract orally administered to

Background: Ulcerative colitis is a chronic inflammatory condition characterized by relapsing and remitting episodes of inflammation limited to the mucosal layer of the colon.Loop ileostomy might be helpful in the management of the disease.

Objective: To evaluate the role of loop ileostomy in the management of ulcerative colitis.

Methods: Between Januarys 2008 and January 2018, patients with fulminant ulcerative colitis resistant to medical therapy had been referred to our surgical unit and were included in our study. Assessment of the patients’ medical status (serum electrolytes, total serum protein, complete blood count, erythrocyte sedimentation rate, general stool exam, and abdominal ultrasound

Evaluation of the Possible Immuno-Protective Effect of Nigella Sativa Seed Oil on Cyclophosphamide- Induced Myelosuppression in Mice
...Show More Authors

Myelosuppression is one of the serious adverse effects of cancer chemotherapy that lead to life threatening febrile neutropenia and considered a limiting factor for successful therapy. Cyclophosphamide a widely used anticancer drugs, induces severe bone marrow suppression by damaging hematopoietic stem cells. As cancer incidence expands globally, the demand for an effective myeloprotective therapy during cancer treatment is also increasing.Nigella sativa seed oil, a well-known plant extract that widely used for various health conditions. This study aims to evaluate the myeloprotective activity of Nigella sativa seed oil in cyclophosphamide-induced myelosuppression mice model. The protective effect of iraqi Juniperus oxycedrus plant on acute kidney injury induced by lipopolysaccharide in mice model
...Show More Authors

Inflammatory control is essential to diminish injury and make renal injury treatment simpler. Proposed therapeutics have primarily targeted pro-inflammatory variables. Juniperus oxycedrus was frequently used to treat a variety of infectious disorders, hyperglycemia, obesity, TB, bronchitis, inflammation, and pneumonia. Juniperus oxycedrus twigs and leaves were defatted with n-hexane using Soxhlet apparatus then the residue of plant material dried and re-extracted sequentially by two different solvents Ethylacetate and methanol. The pro-inflammatory markers IL-1 and iNOS, as well as the potential kidney biomarker KIM-1, TNF-α, and transcription factor NF-KB were measured using the RealTime Quantitative qPCR method. The results showed that J
