1, 3, 4-OXADIAZOLE: Synthesis Derivatives &Biological Activities. A Review article

Heterocyclic compounds serve as crucial structural elements in the field of pharmaceutical medicinal chemistry. The five-membered heterocyclic nucleus of 1,3,4-oxadiazole occupies a distinctive position within medicinal chemistry and plays a vital role in the development of anticancer agents. Recognized as a significant pharmacophore for approximately 85 years, 1,3,4-oxadiazole is in high demand across various biological and chemical disciplines. This small and straightforward nucleus is found in numerous compounds that are the focus of research concerning their properties, synthesis, derivatives, and pharmacological activities, including anticancer, antibacterial, antimalarial, anti-inflammatory, antidepressant, analgesic, and antiviral effects. The information regarding these activities presented in this article may prove beneficial to researchers, facilitating the discovery of new therapeutic agents for the benefit of society.

Introduction:

Allosteric inhibition of EGFR tyrosine kinase (TK) is currently among the most attractive approaches for designing and developing anti-cancer drugs to avoid chemoresistance exhibited by clinically approved ATP-competitive inhibitors. The current work aimed to synthesize new biphenyl-containing derivatives that were predicted to act as EGFR TK allosteric site inhibitors based on molecular docking studies.

Two new organotin(IV) complexes Me2Snesc (C1) and Bu2Snesc (C2) have been synthesised from the reaction of the corresponding organotin(IV) chloride with the Schiff base ligand 3,4-dihydroxybenzaldehyde-4-ethylsemicarbazone (H2esc). The ligand was prepared in two steps. The first step includes the formation of 4-ethylsemicarbazide, which then reacted with 3,4-dihydroxybenzaldehyde to give the title ligand. Complex formation between the organotin(IV) moiety and the anionic form of 3,4-dihydroxybenzaldehy-4-ethylsemicarbazone occurred through the o-dihydroxy positions. New derivatives of Schiff bases were synthesized from the aldehyde derivative (Ma2) which was produced by reacting the mefenamic acid (Ma) with thionyl chloride to obtain the acid halide derivative (Ma1). Compound (Ma1) was dissolved in DMF and mixed with p-hydroxybenzaldehyde which was previously dissolved with pyridine to obtain the aldehyde derivative (Ma2).
